蝎毒组分Ⅰ对卵巢癌细胞SKOV3生长的抑制作用

Inhibitory effects of fraction Ⅰ from scorpion venom on growth of SKOV3 ovarian carcinoma cells

摘要: 目的:观察蝎毒组分Ⅰ(SV-Ⅰ)对卵巢癌细胞SKOV3生长的抑制作用。方法:卵巢癌细胞SKOV3分为空白对照组和SV-Ⅰ处理组(终浓度分别为200、400、800 mg•L-1),采用MTT法检测SV-Ⅰ对卵巢癌细胞SKOV3生长抑制率,集落形成法检测肿瘤细胞增殖,流式细胞术检测细胞周期和细胞凋亡的变化。结果:200、400和800 mg•L-1-1组, SKOV3细胞集落数分别为57.00±6.16和48.00±4.11,明显低于对照组(84.00±5.29)(P<0.01);与空白对照组比较,400 mg•L-1组SKOV3细胞S期细胞数明显减少(P<0.01),G2+M期细胞明显增多 (P<0.01), 800 mg•L-1组SKOV3细胞G2+M期细胞数明显减少(P<0.01),G1期细胞数有增多趋势;与空白对照组比较,400 和800 mg•L-1组SKOV3凋亡细胞数明显增加(P<0.01)。结论:在一定浓度范围内蝎SV-Ⅰ可明显抑制卵巢癌细胞SKOV3的生长,其机制与SV-Ⅰ抑制SKOV3细胞DNA合成、诱导SKOV3细胞发生G2+M和G1期阻滞及诱导SKOV3细胞凋亡有关。

Abstract: To explore the inhibitory effects of fraction Ⅰ from scorpion venom (SV-Ⅰ) on the growth of SKOV3 ovarian carcinoma cells. MethodsSKOV3 ovarian carcinoma cells were divided into two groups: control group and treatment group with SV-Ⅰ(final concentration as 200,400 and 800 mg•L-1,respectively). MTT method was used to detect the inhibitory rate of tumor cells treated with SV-Ⅰ in vitro and colony forming assay was applied to measure the proliferation of tumor cells. Moreover, flow cytometry was further adopted to observe the changes of cell cycle and apoptosis of SKOV3 cells. Results SV-Ⅰ could inhibit the growth of SKOV3 ovarian carcinoma cells significantly at the concentrations of 200, 400 and 800 mg•L-1 and t he inhibitory rates were 29.87 %,48.11% and 67.77 %, respectively (P<0.01). Furthermore, SV-Ⅰ could inhibit colony forming capacity of SKOV3 ovarian carcinoma cells at 400 and 800 mg•L-1 and the numbers of colony were 57.00±6.16 and 48.00±4.11, respectively (P<0.01). Compared with control, cells in S stage were decreased and cells in G2+M stage were increased obviously (P<0.01) after SV-Ⅰ was given at 400 mg•L-1, while cells in G2+M stage were decreased (P<0.01) and cells in G1 stage had a ascending tread after treated with 800 mg•L-1 SV-Ⅰ. Furthermore, apoptosis could be induced obviously by SV-Ⅰ at 400 and 800 mg•L-1 compared with control(P<0.01). Conclusion SV-Ⅰ could inhibit the proliferation and growth of SKOV3 ovarian carcinoma cells and the changes of cell cycle and apoptosis may be important mechanism of inhibitory effects of SV-Ⅰ.
