摘要/Abstract

摘要：

目的：研究基质金属蛋白酶9（MMP-9）和增殖细胞核抗原（PCNA）在端粒酶途径和端粒替代途径骨肉瘤细胞中的表达，探讨二者在骨肉瘤发生及发展中的作用。方法：应用免疫组织化学染色法对27例骨肉瘤组织和10例正常骨组织中的MMP-9、PCNA、人端粒酶催化亚单位（hTERT）和早幼粒细胞白血病小体（PML）进行定量检测。结果：① MMP-9和PCNA在骨肉瘤组织中的表达阳性率分别为88.89%和85.19%，对照组分别为10%和0%，2组阳性率比较差异均有统计学意义（P<0.001）；② MMP-9在端粒酶途径(hTERT阳性)的骨肉瘤组织中表达(+++)、 (++)、(+)和(－)分别为12、5、1和2例；在端粒替代途径(PML阳性)的骨肉瘤组织中表达 (+++)、(++)、(+)和(－)分别为0、4、2和1例，2组等级资料比较差异有统计学意义（P<0.05）；③ PCNA在端粒酶途径的骨肉瘤组织中表达(+++)、 (++)、(+)和(－)分别为10、6、2和2例；在端粒替代途径的骨肉瘤组织中表达 (+++)、(++)、(+)和(－)分别为0、4、2和1例，2组等级资料比较差异有统计学意义（P<0.05）。结论：MMP-9和PCNA在端粒酶途径和端粒替代途径的骨肉瘤细胞中的表达有明显的差异，端粒酶途径骨肉瘤细胞侵袭转移和增殖能力强于端粒替代途径骨肉瘤。

Abstract:

Objective To study the expressions of matrix metalloproteinases-9(MMP-9) and proliferating cell nuclear antigen(PCNA) in telomerase and alternative lengthening of telomere osteosarcoma,and discuss the effects of MMP-9 and PCNA
in the occurrence and development of osteosarcoma. Methods The expressions of MMP-9,PCNA,human telomerase reverse transcriptasc(hTERT) and promyelocytic leukemia body(PML) were examined using immunohistochemical SP staining method in 2
7 cases of osteosarcoma and 10 cases of normal bone tissues.Results ①The positive rates of MMP-9 and PCNA expressions in osteosarcoma tissues were 88.89% and 85.19% respectively,in normal bone tissues were 10% and 0%,there were significant differences between two groups(P<0.001);② The expressions of MMP-9 in osteosarcoma of telomerase were twelve cases of (+++),five cases of (++),one case of (+),two cases of　(－).The expressions of MMP-9 in osteosarcoma of alternative lengthening of telomere were zero case of(+++),four cases of (++),two cases of (+),one case of (－),there were statistically significant differences between telomerase and telomere alternative osteosarcoma( P<0.05).③ The expressions of PCNA in osteosarcoma of telomerase were ten cases of (+++),six cases of (++),one case of (+),two cases of(－),the expressions of PCNA in osteosarcoma of alternative lengthening of telomere were zero case of (+++),four cases of (++),two cases of (+),one case of (－),there were statistically significant differences between telomerase and telome
re alternative osteosarcoma( P<0.05).Conclusion There are statistically significant differences of the expressions of MMP-9 and PCNA between osteosarcoma of telomerase and telomere alternative osteosarcoma.The abilities of metastasis and invasion and proliferation in telomerase osteosarcoma are stronger than those of alternative lengthening of telomere osteosarcoma.
