Abstract: Objective To investigate the effects of advanced glycation end products (AGEs) and Ginkgo biloba L. on proliferation, cell cycle and transforming growth factor β (TGF-β) expression of bovine retinal pericytes, and observe the protection of Ginkgo biloba L. on pericytes. Methods MTT colorimetric assay, flow cytometry, immunofluorescence were used for analysis. Results Ginkgo biloba L. could promote proliferation of pericytes undergoing AGEs. The Ginkgo biloba L. with concentration of 125~400 mg•L-1 could promote proliferation of pericytes obviously (P<0.01) in a dose-dependent manner. Ginkgo biloba L. could inhibit the effects of AGEs on cell cycle of pericytes and induce apoptosis. Ginkgo biloba L. decreased the number of S-phase cells, increased G2-M phase cells, decreased the number of apoptotic pericytes obviously compared with AGEs (P<0.01). Ginkgo biloba L. also down-regulated TGF-β protein expression induced by AGEs. Conclusion AGEs can damage bovine retinal pericytes through one of the pathways of oxidative stress, and antioxidant Ginkgo biloba L. can reduce the toxic effects of AGEs.
