瘤细胞αv整合素对腺病毒载体转染的影响

摘要: 目的:探讨瘤细胞αv整合素在腺病毒载体转染瘤细胞中的作用。方法:带有报告基因LacZ的5型缺陷性腺病毒(AdCMVLacZ)感染肿瘤细胞,X-gal染色,检测腺病毒对肿瘤细胞的转染率;比较M21黑色素瘤细胞及缺乏αv整合素的M21-L对腺病毒转染的易感性;观察含有RGD序列的合成肽GRGDSP对腺病毒转染的影响。结果:腺病毒对各种人瘤细胞系的转染率是不同的,当腺病毒感染复数量(MOI)为50时,A549细胞的转染率最高,达90.2%,Hela细胞的转染率最低,只有7.6%;各种人瘤细胞达到100%转染所需腺病毒量(MOI)是不同的,A549最少,只需75 MOI。在相同MOI下,M21的转染率高于缺乏αv整合素的M21-L。100%转染所需MOI量,M21为100,M21-L为400。M21经GRGDSP处理后对腺病毒转染的易感性降低。结论:肿瘤细胞对腺病毒转染的易感性与瘤细胞αv整合素有关。

Abstract: To study the effect of integrin αv of tumor cells on adenovirus vector transfection. Methods Tumor cells were transfected by vector AdCMVLacZ. The number of transfected cells was assessed by X-gal staining. The sensitivities of M21 melanona cells and M21-L lacking integrin to adenovirus infection were compared. The blocking effect of GRGDSP -a peptide containing RGD sequence on adenovirus transfection after using GRGDSP to treat tumor cells was observed. Results The transfection efficiencyies of adenovirus to different human tumor cells were different. When multiplicity of infection(MOI) of adenovirus was 50, A549 cells had the highest transfection efficiency (90.2%), the transfection efficiency of Hela cells was the lowest, only 7.6%. On the other hand, different tumor cells needed different MOI of AdCMVLacZ to get 100% transfection efficiency. A549 cells needed 75 MOI. The transfection efficiency of AdCMVLacZ in M21 cells was higher than that in M21-L at same MOI. M21 needed 100 MOI for 100% transfection efficiency, however M21-L needed 400 MOI. After M21 was treated with GRGDSP, the transfection efficiency decreased. Conclusion The sensitivity of tumors to adenovirus transfection is related to integrin of tumor cells.
