碘乙酰胺诱导的大鼠胃炎中神经肽免疫阳性神经纤维的变化

Changes of immunoreactive nerve fibres in rats with iodoacetamide-induced gastritis

摘要: 目的:探讨含不同神经肽的神经纤维在碘乙酰胺诱导的大鼠胃炎中的变化,分析神经纤维与免疫细胞的相互关系。方法:采用碘乙酰胺诱导制备大鼠胃炎模型,以纯净水喂养为对照组,采用免疫组织化学法、免疫细胞化学法定量分析对照组及模型组大鼠胃黏膜中含P物质(SP)、神经肽Y(NPY)及血管活性肠肽(VIP)的神经纤维数量及免疫活性细胞。结果:光学显微镜观察模型组和对照组胃黏膜含SP、NPY及VIP神经纤维分布于黏膜层腺体之间。模型组黏膜含SP及VIP的神经纤维数量较对照组明显增加(P<0.05)。模型组大鼠的胃黏膜可见免疫活性细胞,主要为淋巴细胞、浆细胞和肥大细胞,表明SP、NPY表现出免疫活性,免疫细胞与神经纤维毗邻。结论:胃炎模型制作成功;神经肽参与了胃炎的形成过程及免疫调节。

Abstract: To analyze the changes of different neuropeptide-containning nerve fibres in iodoacetamine-induced gastritis, and the relationship between the nerve fibres and immune cells. Methods The animal models with gastritis were induced by iodoacetamide. Controls received water.Gastric specimens from the control and the model groups were examined by immunohistochemistry and immunocytochemistry and the changes of these fibres containing-Substance P (SP), vasoactive intestinal polypeptide (VIP) and neuropeptide Y (NPY) and immunocompetent cells were analyzed. Results The nerve fibres showing immunoreactivity for SP, VIP and NPY were seen in all layers of the stomach. Nerve fibres containing-SP and VIP increased significantly in model group(P<0.05). Immunocompetent cells could be found in mucosa in model group, which were lymphocytes, plasma cells, and mast cells. Immunocompetent cells were very near the fibres. Conclusion The model with mild iodoacetamide-induced gastritis is built successfully. Neuropeptides participate in the processes of gastritis and might involve in neuroimmunomodulation in gastritis.
