Influence of human α-synuclein A30P mutation in neurogenesis in mouse olfactory bulb

摘要:

目的:探讨人α-突触核蛋白(SNCA)点突变A30P对成体小鼠嗅球(OB)中神经发生的影响,阐明其在退行性神经疾病发生中的作用。方法:以C57BL/6正常小鼠(C57BL)、内源性SNCA敲除小鼠(SNCA-/-)、内源性SNCA敲除与人SNCA A30P点突变敲入小鼠(A30P SNCA-/-)为实验动物,分为C57BL、SNCA-/-和A30P SNCA-/-3组,取小鼠OB冠状冰冻切片,进行形态学观察及磷酸化组蛋白3(PH3)、双皮质素(Dcx)、4’,6-二脒基-2-苯基吲哚(DAPI)免疫荧光染色,分析PH3+细胞数、Dcx+细胞的荧光强度以及DAPI染色后的细胞密度。结果:SNCA-/-和A30P SNCA-/-组小鼠OB突触球层(GL)的细胞密度轻微降低,A30P SNCA-/-组小鼠OB轻度变形,其PH3+增殖性神经元也较其他2组显著减少(P<0.05);SNCA-/-和A30P SNCA-/-组小鼠OB中神经母细胞的Dcx荧光强度轻微增加,3组小鼠OB中DAPI染色后的细胞密度未见明显变化。结论:人SNCA A30P突变能减弱成体小鼠大脑OB中的神经细胞增殖,对成体OB中的神经发生有一定抑制作用。

Abstract:

〗Abstract:Objective To investigate the influence of α-synuclein (SNCA) A30P point mutation in the neurogenesis in the olfactory bulb (OB) of adult mice,and to charify its effect in the pathogenesis of degenerative nervous diseases.Methods Normal C57BL mice,endogenous SNCA knockout mice (SNCA-/-) and SNCA knockout + human SNCA A30P knockin mice (A30P SNCA-/-) were used as experimental animals and were divided into C57BL,SNCA-/-, and A30P SNCA-/- groups.The coronal frozen sections of OB were made and the  morphological changes of  OB were  examined.Fluorescence immunohistochemistry of phosphohiston 3 (PH3),doublecortin (Dcx) and 4',6-diamidino-2-phenylindole dihydrochloride (DAPI) staining were carried out to analyze the number of  PH3+ cells,the fluorescence intensity in Dcx+ neuroblasts and the cell densities after DAPI staining in OB.Results The cell densities in glomerular layer (GL) of OB of the mice in SNCA-/- and A30P SNCA-/- groups were decreased slightly.A slight deformation of OB morphology also occurred in A30P SNCA-/- group and the number of PH3+ cells in A30P SNCA-/- group  was decreased significantly compared with C57BL and SNCA-/- groups (P<0.05);the fluorescence intensities of Dcx+ neuroblasts in OB in SNCA-/- and A30P SNCA-/- groups were increased slightly and no evident changes of the cell densities in OB after DAPI staining was observed.Conclusion Human SNCA A30P point mutation can decrease the neuron proliferation in OB of the adult mouse brain,which suggests it play a certain inhibitory effect on the neurogenesis in adult OB.
