姬松茸粗多糖提高脑老化小鼠学习记忆能力的作用及其机制

摘要:

目的:观察姬松茸粗多糖对D-半乳糖(D-gal)致脑老化模型小鼠行为学、海马神经细胞线粒体膜电位及凋亡的影响,探讨其提高脑老化小鼠学习记忆能力的作用及可能机制。方法:40只昆明种小鼠随机分为正常组、模型组及姬松茸粗多糖低、高剂量组,每组10只。制备D-gal小鼠脑老化模型,采用Morris水迷宫和避暗仪测试小鼠的学习记忆能力,采用流式细胞术分析小鼠海马神经细胞凋亡率和线粒体膜电位的变化。结果:行为学测试,与模型组比较,姬松茸粗多糖组小鼠在水迷宫中心区域游泳距离占其总距离百分比明显提高(P<0.05或P<0.01),在第4象限游泳路程明显增多(P<0.05),躲避电击潜伏期明显延长(P<0.05),进入暗箱的错误次数显著减少(P<0.01);与模型组比较,姬松茸粗多糖组小鼠海马神经细胞线粒体膜电位显著升高(P<0.01),细胞凋亡率则明显下降(P<0.05或P<0.01)。结论:姬松茸粗多糖可增强脑老化小鼠的学习记忆能力,其作用机制与提高脑老化小鼠海马神经细胞线粒体膜电位和抑制细胞凋亡有关。

Abstract:

Abstract:Objective To observe the intervention effect of agaricus blazei polysaccharide (ABP) on ethology,mitochondrial membrane potential and apoptosis of hippocampal neuron in mice with brain aging induced by D-gal and to explore its effect and possible mechanism of improving the learning and memory ability of brain aging mice.Methods 40 Kunming mice were divided into four groups with 10 in each group:control group,aging model group,low dose ABP group and high dose ABP group.Morris water maze and avoiding dark instrument were used to detect the learning and memory ability of aging mice induced by D-gal while flow cytometry was used to analyze the changes of mitochondrial membrane potential and apoptosis.Results In the test of ethology,compared with model group,the percentages of distance in the central area of Morris water maze were obviously increased in ABP groups (P<0.05 or P<0.01),and the distances in the fourth quadrant were obviously decreased (P<0.01).Compared with model group,the mitochondrial membrane potential of hippocampal neuron in ABP groups were increased (P<0.01) and the apoptotic rates were decreased significantly (P<0.05 or P<0.01).Conclusion ABP can strengthen the learning and memory ability of aging mice and the mechanism is related to the increasing of  mitochondrial membrane potential of hippocampal neuron and the inhibition of apoptosis.
