清脑止痛胶囊对硝酸甘油致偏头痛大鼠体内炎性细胞因子表达水平的影响及其意义

Effect of QNZTJN on expression levels of inflammatory cytokines in migraine rats induced by nitroglycerin and its significance

摘要:

目的: 皮下注射硝酸甘油(NTG)制备大鼠偏头痛模型,探讨清脑止痛胶囊对大鼠脑组织和血浆中炎性细胞因子表达的影响及其意义。方法: 90只健康成年雄性Wistar大鼠随机分为空白组,模型组,低(0.35g·kg-1)、中(0.70g·kg-1)、高(1.40g·kg-1)剂量清脑止痛胶囊组和阳性药(正天丸1.8g·kg-1)组,每组15只。连续给药7d后,皮下注射NTG复制偏头痛大鼠模型,观察注射NTG180min内大鼠的挠头次数,4h后取血浆和中脑组织。免疫组织化学法和Western blotting法检测大鼠中脑组织中核因子kappaB(NF-κB)和环氧合酶2(COX-2)表达水平,酶联免疫吸附法检测大鼠血浆和中脑组织中前列环素E2(PGE2)、白细胞介素1β(IL-1β)和肿瘤坏死因子α(TNF-α)水平。结果: 与空白组比较,模型组大鼠挠头次数明显增加(P<0.01),中脑组织中NF-κB和COX-2表达水平明显升高(P<0.01),血浆和中脑组织中PGE2、IL-1β和TNF-α水平明显升高(P<0.05或P<0.01);与模型组比较,中、高剂量清脑止痛胶囊组和阳性药组大鼠挠头次数明显减少(P<0.05或P<0.01),中脑组织中NF-κB和COX-2表达水平明显降低(P<0.05或P<0.01),血浆中PGE2、IL-1β和TNF-α水平明显降低(P<0.05或P<0.01)。结论: 清脑止痛胶囊可以缓解大鼠偏头痛症状,对偏头痛有一定的预防作用,其机制可能与调节神经源性炎症有关联。

Abstract:

Objective: To observe the effect of QingNaoZhiTong JiaoNang(QNZTJN) on the expressions of inflammatory cytokines in midbrain tissue and plasma of nitroglycerin(NGT)-induced migraine rats and its significance. Methods: Atotal of 90 male Wistar rats were randomly divided into control group,model group,low dose(0.35 g·kg-1),middle dose(0.70 g·kg-1),high dose(1.40 g·kg-1) of QNZTJN groups and positive drug (Zhengtian Wan,1.8 g·kg-1) control group(n=15).After treated with drugs for 7 d,the migraine models were induced by injecting of NTG into the rats;the forelimb scratching frequency of the rats during 180 min of NTG injection.The midbrain tissue and plasma were obtained 4 h later.The expression levels of nuckear factor kappa B(NF-κB) and cyclooxygenase 2(COX-2) in brain tissue of the rats were detected by immunohistochemistry method and Western blotting method;the levels of prostaglandin E2(PGE2),interleukin-1 beta(IL-1β), and tumor necrosis factor-α(TNF-α) in the midbrain tissue and plasma of the rats were detected by enzyme immunosorbent assay. Results: Compared with control group,the forelimb scratching frequency of the rats in model group was significantly increased(P<0.01),the levels of NF-κB and COX-2 in midbrain tissue were significantly increased(P<0.01),and the levels of PGE,IL-1β and TNF-a were increased in brain tissue and plasma(P<0.05 or P<0.01).Compared with model group,the forelimb scratching frequencies of the rats in middle and high doses of QNZTJN groups and positive drug control group were significantly decreased(P<0.05 or P<0.01),the levels of NF-κB and COX-2 in brain tissue were also significantly decreased(P<0.05 or P<0.01).The levels of PGE2,IL-1β and TNF-a in plasma were decreased (P<0.05 or P<0.01). Conclusion: QNZTJN can relieve the migraine attacks and prevent migraine partly and its mechanism may be related to regulating the neurogenic inflammation.
