ELK-3蛋白在肝癌组织中的表达及其对肝癌HepG2细胞迁移和侵袭能力的影响

Expression of ELK-3 protein in hepatocellular carcinoma tissue and its effects on migration and invasion abilities of HepG2 cells

摘要: 目的:探讨转录因子ELK-3与肝癌患者临床病理特征的关系及ELK-3对人肝癌HepG2细胞迁移和侵袭能力的影响,并阐明其作用机制。方法:选取80例肝癌患者癌组织及49例癌旁正常组织石蜡标本、18例肝癌患者癌组织及癌旁正常组织新鲜术后标本,采用免疫组织化学法和Western blotting法检测ELK-3蛋白在肝癌及癌旁正常组织中的表达,探讨ELK-3与肝癌患者临床病理特征的关系。选取处于对数生长期的人肝癌HepG2细胞,设立对照组(control-siRNA)和ELK-3干扰组(ELK-3-siRNA),将ELK-3 siRNA转染至HepG2细胞,通过细胞划痕实验及体外Transwell小室实验观察肝癌HepG2细胞的迁移和侵袭能力。结果:石蜡组织标本免疫组织化学染色,与癌旁正常组织比较,肝癌组织中ELK-3蛋白阳性表达率明显升高(P<0.05),且ELK-3蛋白阳性表达率与肝癌患者肿瘤数、TNM分期、Edmondson分级和门静脉浸润有关联(P<0.05)。新鲜术后标本Western blotting法检测,与癌旁正常组织比较,18例肝癌患者肝癌组织中ELK-3蛋白表达水平明显升高(P<0.05)。与对照组比较,ELK-3-siRNA组细胞的迁移距离明显变短(P<0.05),侵袭细胞数明显减少(P<0.05)。结论:ELK-3在肝癌组织中的表达水平明显升高。干扰ELK-3会抑制肝癌细胞的迁移和侵袭,表明ELK-3蛋白可以通过抑制肝癌细胞的迁移和侵袭对其产生影响,从而在肝癌发生发展过程中起重要作用。

Abstract: Objective:To explore the relationships between ELK-3 and the clinicopathologic features of the patients with hepatocellular carcinoma (HCC) and the effects of ELK-3 on the migration and invasion abilities of human HCC HepG2 cells,and to clarify their mechanisms. Methods:The paraffin-embedded sections of 80 cases of carcinoma tissue and 49 cases of para-carcinoma tissue,fresh postoperative specimens of 18 cases of carcinoma tissue and para-carcinoma tissue were selected.The expressions of ELK-3 protein in HCC and para-carcinoma normal tissues were detected by immunohistochemistry and Western blotting method.The human HCC HepG2 cells at logarithmic growth phase were divided into control-siRNA and ELK-3-siRNA groups.ELK-3 siRNA was transfected into the HepG2 cells,and the migration and invasion abilities of HepG2 were observed by wound-healing assay and Transwell assay. Results: The immunohistochemistry results showed that compared with the para-carcinoma tissue,the positive expression rate of ELK-3 in the HCC tissue was significantly increased (P<0.05),and the positive expression rate of ELK-3 was positively related to the tumor number,TNM stage,Edmondson classification and venous infiltration of the patients (P<0.05).The Western blotting results showed that compared with the para-carcinoma tissue,the expression level of ELK-3 protein in the HCC tissue of 18 HCC patients were markedly increased (P<0.05).The migration distance and the number of invasion cells in ELK-3-siRNA group were remarkably shorter and less than those in control-siRNA group (P<0.05). Conclusion: The expression level of ELK-3 in HCC tissue is significantly increased,and ELK-3 interference could inhibit the migration and invasion of HepG2 cells,which shows that ELK-3 could exert important effects on the occurrence and development of liver cancer by inhibiting the migration and invasion of HCC cells.
