磷酸化胞外信号调节激酶在小鼠海马发育过程中的表达及其意义

Expression of phosphorylated extracellular signal-regulated kinase during development of hippocampus in mice

摘要: 目的:观察磷酸化胞外信号调节激酶(pERK)在胚龄(E)18 d以及生后不同日龄(P)小鼠海马不同阶段的表达,探讨其在海马发育中的可能作用。方法:分别取E18 d和P 1、3、7、14、21和28 d小鼠的海马组织,每个时间点均作为观察组,采用免疫组织化学和免疫印迹法、结合图像分析技术检测各时间点海马组织中pERK蛋白表达水平。结果:免疫组织化学检测,pERK蛋白主要在海马齿状回(DG)颗粒细胞层和阿蒙角(CA)锥体细胞层神经元的细胞核中表达。E 18 d,海马DG和CA区pERK表达染色浅,排列稀疏,表达水平很低;E 18 d~P 7 d,pERK染色逐渐加深、密集,pERK表达水平逐渐升高,与其他6个时间点比较,P 7 d时pERK表达水平最高(F=34.537,P<0.01)。P 14~21 d,pERK染色逐渐变浅,密集程度逐渐降低,pERK表达水平逐渐下降(F=28.754,P<0.01);P 28 d时pERK表达水平处于稳定的较低水平(F=6.075,P>0.05)。免疫印迹检测,各时间点均有pERK表达,E 18 d~P 7 d,pERK表达水平逐渐升高,与其他6个时间点比较,P 7 d时pERK表达水平最高(F=33.856,P<0.01);P 14~21 d时pERK表达水平逐渐降低(F=22.627,P<0.01);P 28 d时pERK表达处于稳定的较低水平(F=7.421,P>0.05)。结论:小鼠生后早期阶段,即E 18 d~P 7 d,海马发育明显增速,pERK表达水平也随之逐渐升高,P 7 d达高峰,随后逐渐下降直至稳定,pERK参与小鼠海马发育过程中的细胞增殖。

Abstract: Objective:To investigate the expressions of phosphorylated extracellular signal-regulated kinase (pERK) in the hippocampus of the mice of embryonic(E)18 d and postnatal(P)1,3,7,14,21 and 28 d,and to discuss its possible role during the hippocampus development of the mice. Methods:The hippocampus tissues of the mice of E 18 d,P 1 d,P 3 d,P 7 d,P 14 d,P 21 d and P 28 d were obtained.Each time point was used as observation group. Immunohistochemistry,Western blotting and image analysis technique were used to analyze the expression levels of pERK protein in hippocampus tissue at each time point. Results:The immunohistochemistry results revealed that pERK protein mainly expressed in the nucleus of the nerve cells in the granular layer of dentate gyrus and pyramidal layer of Ammon's horn in hippocampus. In the mice of E 18 d, the expression of pERK in DG and CA regions of hippocampus were slightly stained and arranged sparsely, and the expression level was low;and the pERK was deeply stained and the expression levels of pERK were increased gradually with the development of the hippocampus from E 18d to P 7 d;compared with other six time points, the expression level of pERK of the mice of P 7 d was the highest(F=34.537, P<0.01).From P 14 d to P 21 d,the pERK was slightly stained and the density was decreased;the expression levels of pERK were decreased (F=28.754,P<0.01),and then remained stably on P 28 d(F=6.075, P>0.05). The Western blotting results showed that pERK expressed at each time point,from E18 d to P7 d,the expression levels of pERK were increased gradually; compared with other six time points,the expression level of pERK on P 7 d was the highest (F=33.856, P<0.01).The expression levels of pERK were decreased from P 14 d to P 21 d(F=22.627,P<0.01),and then remained stably on P 28 d(F=7.421,P>0.05). Conclusion:The hippocampus development is increased significantly at the early stage of the mice(E18-P 7d),and the expression levels of pERK are increased gradually in the meanwhile;the expression level peaks on P 7 d, and then it is gradually decreased until a stable level. pERK may take part in the cell proliferation in the development of hippocampus of the mice.
