吗啡对胶质瘤C6细胞RNA聚合酶Ⅱ通用转录因子F基因表达的影响

Effect of morphine on gene expression of RNA polymeraseⅡ general transcription factor TFⅡF in C6 glioma cell

摘要: 目的:检测吗啡对C6胶质瘤细胞RNA合成的影响。 方法:通过RT-PCR方法检测吗啡影响C6胶质瘤细胞RNA聚合酶Ⅱ通用转录因子F(TFⅡF)基因表达的变化。 结果:吗啡作用于C6胶质瘤细胞与相应对照组相比,TFⅡF转录产物均明显减少;纳络酮能阻断吗啡对TFⅡF基因表达抑制的作用。结论:吗啡通过μ受体途径介导C6胶质瘤TFⅡF基因表达抑制的作用。

Abstract: Objective To determine the effect of morphine on RNA synthesis of C6 glioma cells. Methods Gene expression level of TFⅡF was detected by RT-PCR. Results Exposure of morphine to C6 glioma cells caused a decrease of TFⅡF gene expression as compared with corresponding control cells, and the effect could be reversed by naloxone. Conclusion The down-regulation of morphine on gene expression of TFⅡF may be mediated by μreceptor.
