高糖对视网膜Müller细胞碱性成纤维细胞生长因子表达及钙离子通道的影响

摘要: 目的:观察在高浓度葡萄糖条件下体外培养兔视网膜Müller细胞碱性成纤维细胞生长因子(basic fibroblast growth factor, bFGF)的表达及其钙(Ca2+)通道的变化。方法:采用免疫细胞化学及透射电镜方法鉴定体外培养的兔视网膜Müller细胞;应用免疫细胞化学方法定性观察高糖条件下视网膜Müller细胞bFGF表达的变化;利用膜片钳技术观察高糖条件下Müller细胞Ca2+通道的变化。结果:高糖可以刺激视网膜Müller细胞表达bFGF,对Ca2+通道无明显影响。 结论:高糖刺激Müller细胞bFGF的表达,而发挥促血管生成的作用,在此过程中Ca2+通道无明显变化。

Abstract: Objective To observe the effects of high concentration glucose on bFGF expression and calcium channels of rabbit retinal Müller cells cultured in vitro. Methods Transmission electron microscopy and immunocytochemical staining method were used to identify Müller cells. The changes of bFGF expressed by Müller cells were studied with immunocytochemical method in a qualitative way. Patch-clamp technique was used to observe the changes of calcium channels caused by high concentration glucose. Results bFGF expression was enhanced obviously by high concentration glucose. Obvious changes of calcium channels were not found. Conclusion bFGF expression secreted by cultured Müller cells in vitro can be enhanced at high concentration glucose. Müller cells play a potential role in neovascularization of diabetic retinopathy. Obvious changes of calcium channels are not found during the process.
