溶栓素抗凝血和抗血栓形成作用的实验研究

摘要: 目的:评价溶栓素的生化药效,研究溶栓素的抗凝血和抗血栓作用。方法:应用玻片法和毛细管法研究溶栓素对小鼠凝血时间的影响;应用大鼠体内、体外血栓形成法及家兔体外血栓形成法研究溶栓素对血栓形成的影响;研究溶栓素对大鼠凝血酶原时间(PT)、活化部分凝血活酶时间(APTT)和纤维蛋白原含量(Fbg)的影响。结果:溶栓素剂量在0.25、0.5及1.0 mg•kg-1体重时,能明显地延长小鼠凝血时间;在0.2、0.4及0.8 mg•kg-1体重时,能有效抑制大鼠体内、体外血栓形成;在0.2 mg•kg-1体重时,能有效地抑制家兔体外血栓形成;在0.2、0.4及0.8 mg•kg-1体重时,能显著地降低大鼠Fbg含量,对APTT、PT未见显著影响。结论:溶栓素具有明显的抗凝血和抗血栓作用,是一种有临床应用前景的治疗血栓性疾病的药物。

Abstract: Objective To study the anticoagulability and antithrombotic activity of the thrombolysin to evaluate its biochmical pharmacodynamics. Methods The effects of thrombolysin on coagulation time in mice were observed by slide method and capillary method. The effects of thrombolysin on antithrombus formation were observed by thrombosis experiment in rats and rabbits. Its effects on prothrombin time (PT),activated partial thormboplastin time (APTT) and the fibrinogen (Fbg) contents were also measured by reagent kits in rats. Results At the dose of 0.25,0.5 and 1.0 mg•kg-1,it could significantly prolong the coagulation time in mice;At 0.2,0.4, and 0.8 mg•kg-1 , thrombolysin inhibited thrombosis of rats in vivo and in vitro. At 0.2 mg•kg-1,it inhibited thrombosis of rabbits in vitro. At the dose of 0.2,0.4,and 0.8 mg•kg-1, it could decrease the Fbg content in rats,and had no effects on PT and APTT. Conclusion The thrombolysin possesses anticoagulability and antithrombotic activity, and it may be a promising medicine for diagnosis of thrombotic diseases.
