红景天对大鼠嗅球中凋亡相关基因表达的影响

AEffects of rhidosin on expression of genes relatedwith apoptosis in olfactory bulbs of senile rats

摘要: 目的:明确红景天素对老龄大鼠嗅球中凋亡相关基因Bcl-2和Bax表达的影响并探讨其意义。方法:健康老龄大鼠20只,随机分成注射红景天素实验组(10只)和等量生理盐水对照组(10只)。断头处死取嗅球,石蜡包埋,连续切片,免疫组化染色,光镜观察。结果:Bcl-2和Bax主要表达于大鼠嗅球中的僧帽细胞。红景天素实验组Bcl-2阳性细胞表达率显著高于生理盐水对照组(P<0.01);Bax阳性细胞表达率实验组虽略低于对照组,但差异无显著性(P>0.05)。 结论:红景天能够诱导老龄大鼠嗅球中Bcl-2蛋白表达增多,抑制僧帽细胞的凋亡,对老龄大鼠嗅球具有抗凋亡作用。

Abstract: Objective To discuss the effects rhidosin in expression of Bcl-2 and Bax genes related with apoptosis in olfactory bulbs of senile rats and its significance. Methods Twenty healthy rats were randomly diveded into rhidosin injected group (n=10) and physiological saline control group. After decaptation the olfactory bulbs were immediately injected with neutral formalin, followed with paraffin-embedding, serial sectioning, immunohistochemical staining and light microscopic observation. Results Bcl-2 and Bax mainly expressed on mitral cells of olfactory bulbs in senile rats. The positive expression rate of Bcl-2 gene in the rhidosin group was significantly higher than that in the physiological saline control group (P<0.01); the positive expression rate of Bax gene in the rhidosin group was slightly lower than that in the physiological saline control group (P>0.05). Conclusion Rhidosin can increase the expression of Bcl-2 protein in olfactory bulbs of senile rats, inhibit the apoptosis of mitral cells and has the effect of antiapoptosis in olfactory bulbs of senile rats.
