摘要：

目的：探讨大鼠发生心肌缺血性纤维化后转录因子（NF-κB）的表达及作用，阐明其与心肌缺血性纤维化发生发展的关系。方法：20只Wistar雄性大鼠随机分为正常对照组、盐酸异丙肾上腺素注射液(Iso)12 h组、Iso 72 h组和Iso 1周组，每组5只。除正常对照组外,其他各组大鼠皮下均注射 Iso (2 mg /kg体质量)。按对应时间点取心脏组织，采用常规染色，免疫组织化学染色方法分析NF-κB蛋白表达。结果：HE染色结果显示正常对照组心肌排列整齐，横纹清晰，胞核明显，无细胞肿胀。注射Iso　12 h后，心肌出现较多点状坏死。注射Iso 1周后，出现界限清晰、多发散在坏死灶。Masson三色染色结果显示正常对照组胶原仅有少量表达，而注射Iso 12　h后，胶原量明显增加(P<0.01)，直至1周达到高峰。NF-κB免疫组织化学染色结果显示正常对照组中可见呈浅黄色或综黄色阳性物质。注射Iso 12 h后，NF-κB阳性物质表达明显增加。注射1周后，NF-κB阳性表达达到最大值(P<0.05)。结论：NF-κB表达随Iso注射时间的延长而明显增加，提示NF-κB参与了大鼠心肌坏死后纤维化的发生及发展过程。

Abstract:

Abstract:Objective
To investigate the expression and role of NF-κB in rat's with myocardial ischemic fibrosis,and illuminate the relations between NF-κB and occurrence and development of myocardial ischemic fibrosis. Mthods 20 male Wistar rats were randomly divided into control group，model group(12 h,72 h and 1 week)(n=5)， Except control group,all the rats in the other groups were treated with isoproterenol hydrochloride(2 mg?kg-1).Conventional staining,immunohistochemical staining and Masson staining were used to analyze expression of NF-κB protein in heart tissues. Results The results of HE staining showed the myocardium tissue had been a good order,the clear nuclear arms,no cell swollen. The mycardium presented many punctual necrosis at 12 h. The myocardium appeared clear boundary and multi-radiations necrosis stove at 1 week. The results of Masson staining showed the myocardium had only a small number of collagen in normal control group,the expression of collagen was obviously increased at 12 h(P<0.01) and reached the arrived peak at 1 week. The results of NF-κB protein expression showed there was positive matter of buff or yellow. The NF-κB expression was increased obviously at 12 h and reached the peak at 1 week(P<0.05). Conclusion Along with,The NF-κB expression is increased obviously which indicats that NF-κB is involved in the occurrence and development of myocardial ischenic frbrosis in rats.with the prolongation of Iso injection time.
