选择性COX-2抑制剂对角膜移植术后新生血管形成的抑制作用

Inhibitory effects of cyclooxygenase-2-selectiveinhibitor on angiogenesis postkeratoplasty

摘要: 目的:探讨选择性环氧化酶-2(COX-2)抑制剂在角膜移植术后新生血管形成中的抑制作用。方法:SD大鼠及Wistar大鼠各20只,在SD大鼠与Wistar大鼠之间建立角膜移植动物模型,分为选择性环氧化酶-1(COX-1)抑制剂小剂量阿司匹林给药组、选择性COX-2抑制剂西乐葆给药组、非选择性COX抑制剂消炎痛给药组及生理盐水阴性对照组。对角膜移植术后角膜混浊、水肿、新生血管形成情况评分得出角膜排斥反应指数(RV),对角膜移植术后新生血管形态计算得出新生血管反应区域。并进行角膜病理切片检查。结果:选择性COX-1抑制剂组及阴性对照组大鼠在7 d时RV值>5,排斥反应已经发生,选择性COX-2抑制剂组及非选择性COX抑制剂组未发生排斥反应。裂隙灯检查及角膜病理切片检查可见选择性COX-2抑制剂组及非选择性COX抑制剂组术后7d出现植床周围少许新生血管。选择性COX-1抑制剂组及阴性对照组术后7 d时新生血管接近创口。选择性COX-2抑制剂组及非选择COX抑制剂组角膜新生血管反应区域及RV值均少于选择性COX-1抑制剂组及阴性对照组。结论:COX-2选择性抑制剂可抑制角膜移植术后新生血管的产生。

Abstract: Objective To study the inhibitory effects of cyclooxygenase-2-selective inhibitor on angiogenesis postkeratoplasty. Methods The rat keratoplasty models between SD and Wistar rats were made. All the rats were taken orally cyclooxygenase-1-selective inhibitor,cyclooxygenase-2-selective inhibitor,non selective-cyclooxygenase inhibitor, and physiological saline separately. The rejection values (RV) according to edema,opacity and new vessel of the cornea graft were measured. The new vessel response area was calculated by observing the new vessel of cornea. Results The RV values of cyclooxygenase-1-selective inhibitor and control groups were more than 5 at the 7 day after operation. Rejection had been found in these two groups while it hadn′t in other two groups. The new vessel of cyclooxygenase-2-selective inhibitor group and cyclooxygenase-1-selective inhibitor group at the 7 day could be detected by slit lamp on the cornea bed. The new vessel of other groups approached the cornea graft. The pathological section examination had the same conclusion. The value of RV and the new vessel response area of the groups in which the rats took cyclooxygenase-2-selective inhibitor and non selective-cyclooxygenase inhibitor were less than those of other groups. Conclusion Cyclooxygenase-2-selective inhibitor can inhibit angiogenesis postkeratoplasty.
