表皮生长因子及胶原网架对培养成纤维细胞的影响

Effects of EGF and collagen scaffold on cultureddermal fibroblasts in vitro

摘要: 目的:观察表皮生长因子(EGF)及胶原网架对体外培养的皮肤成纤维细胞(FB)增殖的影响。方法:酶消化法分离大鼠皮肤FB,以含有10%新生牛血清的DMEM培养液进行培养,采用细胞计数、MTT法、组织学方法测定细胞增殖能力和形态变化。结果:培养液中EGF浓度大于5 μg•L-1时,FB的增殖活性随EGF浓度的增加而增加,在 20 μg•L-1浓度时达到最大。植入胶原网架中的FB,随着培养时间的延长,细胞数量增加缓慢。结论:EGF对FB的增殖具有促进作用,而胶原网架对FB的增殖速率具有一定的调节作用。

Abstract: Objective To investigate the effects of EGF and collagen scaffold on cultured rat dermal fibroblasts in vitro. Methods The fibroblasts were isolated by enzyme digestion and cultured with DMEM medium contained new bovine serum (10%). The proliferation abilities of the fibroblasts were evaluated by MTT assay and the morphologic changes of the fibroblasts were evaluated by histological method.Results When the EGF concentration was above 5 μg•L-1, the proliferation abilities of cultured fibroblasts increased markedly. The number of cultured fibroblasts planted into collagen scaffold increased slowly. Conclusion EGF has the auxoaction on proliferation of the cultured fibroblasts in vitro. Collagen scaffold has some regulatory effects on proliferation rate of the cultured fibroblasts in vitro.
