Effect of Compound Angelica Injection on expression of brain derived neurotrophic factor of rats with cerebral ischemia injury

Abstract:

Objective To investigate the effect of Compound Angelica Injection (CAI) on the expressions of brain derived neurotrophic factor (BDNF) in brain tissue and serum in the rats with cerebral ischemia injury, and to explore the related mechanism of effect of CAI on cerebral ischemia injury. Methods 50 SD rats were randomly divided into sham operation group (n=10) and cerebral ischemia injury group (n=40), and the rats in cerebral ischemia injury group were reproduced by middle cerebral artery occlusion (MCAO). Thirty successfully modeling rats were randomly divided into model group, CAI group and edaravone group (n=10). The neurologic scores of the rats after waking up were measured. The pathomorphology of brain tissue of the rats was detected by HE staining 7 d later. The levels of BDNF in the brain tissue and serum of the rats were detected. Results Compared with sham operation group, the neurologic score of the rats in model group was increased (P<0.05); the neurocytes of rats were changed to swell, necrosis, and interstitial edema; the levels of BDNF in brain tissue and serum showed a tendency of increasing; the level of BDNF in brain tissue of the rats in CAI group was increased significantly (P<0.05), and the serum BDNF level was a little higher. Compared with model group, the neurologic score of the rats in CAI group was decreased (P<0.05); the neurocytes of rats were mild edema; the levels of BDNF in brain tissue and serum showed a tendency of increasing. Conclusion CAI may has a positive function in increasing BDNF expression,and the mechanism may be related to the stress reaction of corresponding neural factors in brain tissue under ischemia injury condition.
