Effect of Compound Angelica Injection on expression of brain derived neurotrophic factor of rats with cerebral ischemia injury

Objective To investigate the effect of Compound Angelica Injection (CAI) on the expressions of brain derived neurotrophic factor (BDNF) in brain tissue and serum in the rats with cerebral ischemia injury, and to explore the related mechanism of effect of CAI on cerebral ischemia injury. Methods 50 SD rats were randomly divided into sham operation group (n=10) and cerebral ischemia injury group (n=40), and the rats in cerebral ischemia injury group were reproduced by middle cerebral artery occlusion (MCAO). Thirty successfully modeling rats were randomly divided into model group, CAI group and edaravone group (n=10). The neurologic scores of the rats after waking up were measured. The pathomorphology of brain tissue of the rats was detected by HE staining 7 d later. The levels of BDNF in the brain tissue and serum of the rats were detected. Results Compared with sham operation group, the neurologic score of the rats in model group was increased (P<0.05); the neurocytes of rats were changed to swell, necrosis, and interstitial edema; the levels of BDNF in brain tissue and serum showed a tendency of increasing; the level of BDNF in brain tissue of the rats in CAI group was increased significantly (P<0.05), and the serum BDNF level was a little higher. Compared with model group, the neurologic score of the rats in CAI group was decreased (P<0.05); the neurocytes of rats were mild edema; the levels of BDNF in brain tissue and serum showed a tendency of increasing. Conclusion CAI may has a positive function in increasing BDNF expression,and the mechanism may be related to the stress reaction of corresponding neural factors in brain tissue under ischemia injury condition.

Influence of irreversible electroporation mediated HPV16 E6 shRNA interference plasmid in proliferation of cervical cancer SiHa cells

Objective To explore the feasibility of using irreversible electroporation(IRE) mediating HPV16 E6 shRNA into cervical cancer cell line SiHa,and to clarify the influence of their co-effect on the proliferation of SiHa cells and its mechanism. Methods A HPV16 E6 gene specific interference sequence was inserted in pGenesil-1 to build a interference vector.10 pulses of IRE with 800 V,100 μs, and 1 Hz were applied to the suspension of SiHa cells and vectors.According to the treatment factors,control group,IRE group,pGenesil-N group,pGenesil-N+ IRE group,pGenesil-E6 group and pGenesil-E6+IRE group were set up.The expression of green fluorescent protein (GFP) and transfection efficiency were confirmed by inverted fluorescence microscope 24 h after the vector was transfected by IRE,and the expression efficancy of GFP was calculated.The expression levels of E6 mRNA and protein were detected by RT-PCR and Western blotting method which was also applied to detect the expressions of P53 and PCNA.The proliferative activity of SiHa cells was determined by CCK-8 assay. Results Enzyme digestion and DNA sequencing verified that the vectors were correctly constructed. GFP was seen under inverted fluorescence microscope 24 h after IRE transfection.Compared with IRE group,the expression levels of E6 mRNA and protein were decreased detected by RT-PCR and Western blotting method after the vectors were treated with IRE,the P53 protein expression level was increased(P<0.05),and the PCNA expression level was decreased(P<0.05). The CCK-8 assay results showed the proliferative activity of SiHa cells in pGenesil-E6+IRE group was decreased more obviously than that in pGenesil E6 group (P<0.05). Conclusion IRE can play the role of gene transfection of mediating HPV16 E6 shRNA into SiHa cells,and their co-effect can significantly inhibit the proliferation of SiHa cells.

Protective effect of phenylephrine on irradiated injury of osteoblasts in rats

Objective To investigate the effect of phenylephrine on the morphology and the expression level of Nampt mRNA in the irradiated osteoblasts of the rats,and to clarify the possible protective effects of phenylephrine on this injury. Methods The osteoblasts cultured by conventional tissue were randomly divided into control group, phenylephrine group,simple irradiation group,and phenylephrine+irradiation group.The cells in phenylephrine group were treated with 100 mmol·L-1 phenylephrine 30 min before irradiation.The cells in simple irradiation group were irradiated with 8 Gy X-ray.The osteoblasts were observed by microscope and were collected at the time points of 8,16,32 and 64 h after irradiation.The Nampt mRNA expression level in osteoblasts was examined by RT-PCR. Results There was no obviously morphological and number changes of osteoblasts in phenylephrine group compared with control group.The osteoblasts in simple irradiation group were atrophied,especially at 8 h; the number of osteoblasts in simple irradiation group was 0.82,0.37,0.24 and 0.21 times as control group respectively at 8,16,32 and 64 h. Atrophy of osteoblasts was alleviated in phenylephrine+irradiation group; at 8,16,32 and 64 h,the number of osteoblasts was 0.91,0.83,0.72 and 0.75 times as control group,and was 1.09,2.24,3.00 and 3.60 times as simple irradiation group.The expression level of Nampt mRNA in the osteoblasts in simple irradiation group was reduced compared with control group(P<0.01)at 8,16,32 and 64 h. There were significant differernces of the expression levels of Nampt mRNA in the osteoblasts between phenylephrine+irradiation group and control group(P<0.01),and there were significant differernces between phenylephrine+irradiation group and simple irradiation group(P<0.01)at the different time points. Conclusion Phenylephrine could reduce the atrophy of osteoblasts in the rats induced by irradiationand up-regulate the expression level of Nampt mRNA.Its protective effect on irradiated injury may be related to the up-regulation of the expression level of Nampt mRNA.

Induction effect of TTF1-NP on human hepatoma cell apoptosis through ERS-mediated pathway

Objective To explore the effects of different doses of 5,2',4'-trihydroxy-6,7,5'-trimethoxyflavone nanoparticles (TTF1-NP) on theapoptosis of human hepatoma cells and human normal hepatocytes, and to explore their mechanisms through endoplasmic reticulum stress (ERS)-meditated apoptosis pathway. Methods The human hepatoma cell lines (HepG2,Hep3B and PLC/PRF/5) and human hepatocytes (Chang Liver) were used as cell model,and divided into vehicle,5-Fu and TTF1-NP treated groups with the concentrations of 50,100 and 200 μmol·L-1 respectively. The inhibitory effects of TTF1-NP on the cell growth were assessed using MTT assay and the best inhibitory one (HepG-2) was selected as the main research cell lines.Flow cytometry was used to detect the TTF1-NP-induced apoptosis; Western blotting and immunocytochemistry were used to determine the expressions of ERS key proteins.Finally,the expressions of key proteins were detected by Western blotting after using the ERS inhibitor 4-PBA. Results Compared with vehicle group,the inhibitory rates of growth of 4 kinds of human hepatoma cells in different concentrations of TTF1-NP groupswere increased(P<0.05 or P<0.01); moreover,the inhibitory effects of TTF1-NP were in a time-and dose-dependent manner.Compared with vehicle group,the apoptotic rates of the cells in TTF1-NP groups were increased in a dose-dependent manner(P<0.05 or P<0.01); the expression levels of ERS key proteins GRP78 and caspase-4 were increased with the increasing of the concentration of TTF1-NP(P<0.05 or P<0.01).The expression levels of ERS key proteins GRP78 and caspase-4 induced by TTF1-NP were inhibited by ERS inhibitor 4-PBA(P<0.05 or P<0.01). Conclusion TTF1-NP can induce the apoptosis of HepG2 cells; ERS pathway plays a central role in TTF1-NP-induced apoptosis of HepG-2 cells.

Construction and identification of urokinase-type plasminogen activator biosensor plasmid

Objective To construct the eukaryotic expression vector urokinase-type plasminogen activator (uPA) biosensor which was the composition of the fusion protein enhanced cyan fluorescent protein-uPA (substrate)-yellow fluorescent protein variant (ECFP-uPA substrate-linker-YPet). Methods By the template Src-biosensor,the YPet primers were designed by Primer Premier 5.0 software,and the restriction enzyme sites,uPA substrate gene sequence and linker were added in its 5' end.With the intermediate vector pDMTM-18T,an eukaryotic expression vector which contained a fusion protein of ECFP-uPA substrate-linker-YPet was constructed by genetic engineering.Then the uPA biosensor was transfected into 293T cells.The transfection efficiency and expression of fusion proteins were observed after 24 h.Fluorescence resonance energy transfer (FRET) was observed by the inversion fluorescence microscope and measured by the MetaFlour FRET 4.6 software. Results The uPA biosensor vector was confirmed by the fragment of PCR and double restriction enzyme digestion.The transfection efficiency was nearly 40%.The immunofluorescence detection results displayed that uPA biosensor fusion protein expressed in the 293T cells membrane and the FRET of uPA biosensor in the living 293T cells was observed after incubation with the recombinant human uPA(rhuPA). Conclusion uPA biosensor is successfully constructed and it could be used as a molecular probe to study the temporal and spatial variation of uPA in living cells.

Expression of dopamine D1 receptors in hippocampal dentate gyrus in rats with vascular dementia

Objective To observe the extracellular content of dopamine (DA) and expression of D1 receptors in hippocampal dentage gyrus (DG) in the model rats with vascular dementia (VD),and to investigate the relationship between them. Methods 12 male SD rats were randomly divided VD group and sham-operation group,and the VD model was prepared by permanent bilateral carotid occlusion.The extracellular content of DA in the DG was determined by in vivo microdialysis and HPLC,and the expression of D1 receptors was measured by immunehisto-chemistry. Results The DA content in the DG of the rats in VD group was lower than that in sham-operation group (P<0.05).The number of D1 receptor-positive cells in the DG hilus in VD group was increased compared with sham-operation group (P<0.05),whereas the expression of D1 receptor in DG granule cell layer did not change (P>0.05). Conclusion The DA content in the hippocampal DG is decreased in the VD rats,and its function may be compensated by the up-regulation of D1 receptors in the DG hilus.

Biocompatibility and brain-targeting ability of nano-micelles on glioma cells

Objective To investigate thebiocompatibility and safety of nanoscale brain-targeting-carrier micelles[poly(ethylene)-b-poly(lactic acid) /OX26 conjugate micells(copolymer/OX26)],and to explore its possibility as brain-targeted-drug carrier for brain glioma. Methods The C6 glioma cells were cultured in vitro and divided into experimental groups with different concentrations (10,20,40,80 mg·L-1) of nano micelle,and the medium without micelle was used as control group.The inhibitory effect of nano-micelles on the rat brain glioma C6 cells was examined by Trypan blue cell counting assay.Flow cytometry (FCM) was used to detect the changes of apoptosis and cell cycle of C6 cells,and confocal laser scanning microscope (CLSM) was performed to analyze the distribution of copolymer/OX26 into C6 cells. Results The results of Trypan blue cell counting assay showed copolymer/OX26 didn't affect the growth of C6 cells,and there were no significant differences in the number of C6 cells between control group and expreimental groups(P>0.05).The results of FCM showed that the cell cycle and and the apoptotic rates of C6 cells had no changes compared with control group(P>0.05).The results of CLSM showed that the fluorescence intensities in experimental groups were higher than those in blank micelles group and blank control group(P<0.05),and they were increased in dose-and time-dependent manner(P<0.05). Conclusion Copolymer/OX26 has no effect on the growth and apoptosis of glioma cells.By bonding OX26, copolymer/OX26 can significantly increase the intake of C6 cells on the nano micelles.

Effects of ionizing radiation on autophagy and apoptosis in MCF-7 cells with Beclin 1 over-and low-expressions and their regulating mechanisms

Objective To establish the MCF-7 cell models of Beclin 1 over-and low-expressions, and to detect the autophagic and apoptotic changes after 4 Gy irradiation, and to explore their molecular regulation mechanisms. Methods MCF-7,MCF-7+4Gy,MCF-7-Beclin 1+4Gy and MCF-7-Belcin 1 RNAi+4Gy groups were set up. Molecular biology method was used to construct Beclin 1 over-expression vector pcDNA3.1-Beclin 1, and to estabilish the Beclin 1 over-and low-expression cell models. After the cells were irradiated with 4 Gy, the autopahgic cell percentages were measured by fluorescence microscope with MDC staining, the apoptotic cell percentages were measured by FCM with AnnexinⅤ-FITC and PI staining, and the expressions of Beclin1, P53, Bcl-2 and Bax proteins were measured by Western blotting method. Results Compared with MCF-7 group, the autophagic and apoptotic cell percentages in MCF-7+4 Gy, MCF-7 Beclin 1+4 Gy and MCF-7-Beclin 1 RNAi+4 Gy groups were significantly increased (P<0.05 or P<0.001 ), especially in MCF-7 Beclin 1+4 Gy group which was significantly higher than those in MCF-7+4 Gy (P<0.05); while there was significant difference in the necrotic cell percentages between various groups. After 4 Gy irradiation, compared with MCF-7 group, the expression levels of Beclin 1, P53 and Bax proteins in MCF-7+4 Gy and MCF-7-Beclin 1+4 Gy groups were increased, but the expression levels of Bcl-2 protein were decreased, especially in MCF-7-Beclin 1+4 Gy group. Conclusion The MCF-7 cell models of Beclin 1 over-and low-expressions are successfully established, and ionizing radiation could induce the autophagy and apoptosis of MCF-7 cells, which is more obvious in Beclin 1 over-expression MCF-7 cells. Beclin 1 can activate P53, inhibit Bcl-2 and activate Bax,which forms the regulation of autophagy and apoptosis by P53.

Damage of endothelial cells induced by OX-LDL and its influence in expressions of TLR-4 and EpCAM

Objective To study the damage effect of oxidized low density lipoprotein (OX-LDL) on the endothelial cells and its influence in the expressions of Toll like receptor 4 (TLR-4) and epithelial cell adhesion molecule(EpCAM). Methods The human endothelial cells ECV-304 were cultured in vitro and treated by different concentrations of OX-LDL for 24 h, and the cell vitality was measured by Taipan blue rejection test.The reactive oxygen species(ROS) level,mitochondrial membrane potential(MMP),cell cycle and apoptosis were detected by flow cytometry (FCM).The expression of TLR-4 and EpCAM were also analyzed by FCM. Results Compared with control group, the ROS levels in ECV-304 cells in 25,50,100, and 200 mg·L-1 OX-LDL were increased,but the cell vitalities and MMP were decreased(P<0.05). Compared with control group, the percentages of S phase cells in 25 and 50 mg·L-1 OX-LDL groups were increased,the G0G1 phase cells were slightly reduced, and the SubG1 phase cells (apoptotic cells) were increased with the increasing of OX-LDL concentration. Compared with control group, the TLR-4 and EpCAM expression levels in ECV-304 cells in 50 mg·L-1 OX-LDL group at 24 h were significantly increased (P<0.05). Conclusion OX-LDL could increase the ROS level and induce the apoptosis in vascular endothelial cells,while increase the cell vitality and MMP.During the damage process, the expresion levels of TLR-4 and EpCAM are up-regulated.

Role of miR-122 in bone marrow derived stem cell therapy for acute liver injuryin rats

Objective To investigatet the effects of miR-122 in the therapy of bone marrow derived stem cells (BMSCs) for acute liver injury in the rats, and to clarify the mechanism. Methods The BMSCs were isolated from the bone marrow of male rats by density gradient centrifugation. The BMSCs were divided into transfection group and control group. The BMSCs in transfection group were transfected with miR-122 mimics by liposome, while the BMSCs in control group were not. 60 SD rats with acute liver injury induced by 10%CCl4 were randomly divided into control group (the saline was injected through mainline), normal treatment group (the normal BMSCs were injected through mainline) and experimental therapy group (the BMSCs transfected with miR-122 mimics by liposome were injected through mainline) (n=20).The liver function and tissue pathology were examined at 1 d, 7 d and 14 d after transplantation. Results The expression level of ALB in BMSCs was up-regulated, while the AFP expression level was down-regulated after the transfection of miR-122 mimics. At 1 d after transfection of BMSCs, the serum alanine aminotransferase(ALT) and aspartate aminotransferase(AST) activities had no significant difference between normal treatment group and experimental therapy group. At 7 d and 14 d after transfection of BMSCs, the serum ALT and AST activities in experimental therapy group were obviously lower than those in normal treatment group (P<0.05). The liver congestion,cytoplasm degeneration and liver cell necrosis in experimental therapy group were improved compared with normal treatment group. Conclusion The up-regulation of miR-122 expression in BMSCs would promote its differentiation into hepatocyte like cells, which plays a role in promoting the recovery of liver injury.

Protective effect of FTY720 on hepatic injury in experimental hepatic fibrosis mice

Objective To observe the protective effects of FTY720 on the Con A-induced mouse hepatic fibrosis injury, and to find the possible mechanisms of protective effects. Methods The pathologic models of hepatic fibrosis injury in the mice caused by Con A were set up.Forty mice were randomly divided into control group, model group, high dose of FTY720(4 mg·kg-1) group and low dose of FTY720 (1 mg·kg-1) dose group (n=10). The serum alanine aminotransferase(ALT) and asparate aminotransferase (AST) activities, hepatic index and pathological changes of hepatic tissue were detected. Results Compared with model group, the serum ALT and AST activities in low and high doses of FTY720 groups were decreased significantly (P< 0.05 or P< 0.01). The optical microscope results showed that there were inflammatory cells and hepatocellular necrosis in model group. The masson staining results showed that there were surrounding fiber bundle and hepatic lobule fusion in model group; compared with model group, the damage degree in low and high doses of FTY720 groups was reduced. The protective effects of FTY720 on hepatic injury showed linear relation to the drug dose. Conclusion FTY720 could decrease the levels of ALT/AST, thus FTY720 alleviate hepatic damage degree and delay the process of hepatic fibrosis.The protective effects of FTY720 on hepatic injury in experimental hepatic fibrosis mice may be related to the mechanisms mentioned above.

Induction effect of baicalin on apoptosis of human colon cancer SW480 cells and its mechanism

Objective To investigate the influence of baicalin in human colon cancer SW480 cells,and to clarify its mechanism. Methods The SW480 cells were cultured and divided into blank control and 25,50 and 100 μmol·L-1 baicalin groups.The proliferation activity was detected with CCK-8 assay.The morphological changes of SW480 cells were detected by Annexin Ⅴ-FITC and DAPI coloration.The protein expression levels of Bcl-2,caspase 3 and caspase 9 were detected by Western blotting method. Results The CCK-8 assay results showed that the proliferation activities of SW480 cells in 25,50 and 100 μmol·L-1 baicalin groups were decreased significantly compared with blank control group at the time points of 24 h,48 h and 72 h (P<0.01),the proliferation activities of SW480 cells in 25 μmol·L-1 baicalin groups were decreased significantly compared with blank control group at the time points of 48 and 72 h (P<0.01).Cell shrinkage and nucleus fragmentation were observed in the SW480 cells after treated with 50 μmol·L-1 baicalin for 48 h.The Western blotting assay results showed that compared with blank control group,the protein expression levels of caspase 3 and caspase 9 in 25,50 and 100 μmol·L-1 baicalin groups were increased significantly (P<0.05 or P<0.01),and the protein expression levels of Bcl 2 in 25,50 and 100 μmol·L-1 baicalin groups were decreased significantly (P<0.05 or P<0.01). Conclusion Baicalin can induce the apoptosis in SW480 cells,and the effect might be involved with the mitochondrial apoptotic pathway.

Objective Using serum pharmacology method to assess the optimal dose of Rhodiolasachalinensis extract(RSE)and Codonopsispilosula extract(CPE)combination as well as the effects of the combination on the immune function of mice. Methods Using the 3×3 facterial design,nine medicated serum of CPE and RSE mixture were made to assess the optimum combinational dose of CPE and RSE by detecting the T,B lymphocyte proliferation abilities and NK cell activity in vitro.Using the optimum combinational dose and reducing 2.5,5 and 10 times as high,middle and low doses of RSE+CPE groups,and the T,B lymphocyte proliferation abilities and the activity of NK cells in the mice were detected in vivo. Results The result of serum pharmacology indicates that compared with control group,the proliferation abilities of T,B lymphocytes and the activity of NK cells inducing by ConA and LPS in 200 mg·kg-1 RSE +790 mg·kg-1 CPE group were increased (P<0.05).The result of in vivo experiment indicated that compared with cyclophosphamide group,the spleen indexes(SI) in middle and high doses of RSE+CPE groups were significantly increased (P<0.05); compared with low dose of RSE+CPE group,the WBC number in middle dose of RSE+CPEgroup was significantly increased (P<0.05).Compared with cyclophosphamide group,the T and B cell proliferation abilities induced with ConA and LPS and killing activities of NK cells in low,middle and high doses of RSE+CPE groups and positive drug CVT-200 group were significantly increased (P<0.05).Compared with CVT-200 group,low and high doses of RSE+CPE groups,the T and B cell proliferation abilities induced with ConA and LPS and the activity of NK cells in middle dose of RSE+CPE group were significantly increased(P<0.05). Conclusion RSE and CPEmixture can enhance the immune function of mice; RES 200 mg·kg-1 and CPE 790 mg·kg-1 are the optimal doses.

Objective To investigate the protective effects of rhein lysinate (RHL) on the blood vessel damage induced by oxidative stress in the mice,and to explore its mechanism. Methods The mouse models of oxidative damage were established by intraperitoneal injection of paraquat.30 C57 mice were randomly divided into control,paraquat model,and RHL prevention groups.The mice in RHL prevention group were given RHL by gavage for one week before performing model.The mice in other two groups were given equal volume of distilled water.For making model,paraquat was intraperitoneally injected in the mice in paraquat model and RHL prevention groups once a week for two weeks.The activities of superoxide dismutase (SOD) and glutathione peroxidase(GSH-Px) and the content of serum malonaldehyde (MDA) of the mice were detected 2 weeks after modeling.The pathological profile of blood vessel was observed by hematoxylin and eosin (HE) staining and the level of reactive oxygen species was observed by DCFH-DA staining.The expressions of genes related to blood vessel damage were detected by Western blotting method. Results Compared with control group,the activities of SOD and GSH-Px were decreased and the content of MDA was increased in paraquat model group (P<0.05).Compared with paraquat model group,the activities of SOD and GSH-Px were increased and the content of MDA was decreased in RHL prevention group (P<0.05).The pathological examination indicated the structure of blood vessel of the mice was damaged and the level of reactive oxygen species of blood vessel was increased (P<0.05) in paraquat model group.The pathological changes were significantly improved and the level of reactive oxygen species of blood vessel of the mice was decreased (P<0.05) in RHL prevention group.The Western blotting analysis showed that compared with control group,the expression levels of nitric oxide endothelial synthase (eNOS) and caspase-3 of the mice in paraquat model group were decreased (P<0.05),however the expression level of cleaved fragment of caspase-3 was increased (P<0.05).Compared with paraquat model group,the expression levels of eNOS and caspase-3 of the mice in RHL prevention group were increased (P<0.05) and the expression level of cleaved fragment of caspase-3 was decreased (P<0.05). Conclusion Paraquat could induce vascular cell damage in vivo through increasing the levels of reactive oxygen species,and RHL could antagonize the effects of paraquat by scavenging reactive oxygen species,and up-regulating the eNOS expression and reducing the expression of the cleaved fragment of caspase-3.

Expressions of TLR4 and Dectin-1 in cornea tissue of rats with Aspergillus fungal keratitis and their effects in pathogenesis of keratitis

Objective To explore the expressions of Dectin-1 and Toll-like receptor 4 (TLR4) and the secretory charateristics of inflammatory factors of the Aspergillus fumigatus infected cornea of the rats,and to clarify theirs role in the pathogenesis of rat keratitis. Methods 40 SD rats were separated into control group(n=10) and aspergillus keratitis group(n=30).The rats in aspergillus keratitis group were infected by Aspergillus fumigatus in two eyes for 12,24 and 48 h,and the cornea tissue was took and ELISA kit was used to detect the level of inflammatory cytokines IL-1,TNF-a,IL-6,IL-10,and NF-κB; immunohistochemistry,Western blotting and RT-PCR methods were used to determine the Dectin-1 and TLR4 protein and mRNA expressions. Results The rat models were successfully established.The eyes of the rats in control group were limpid tested by corneal slit lamp,however,there existed a clear boundary film on the surface of eyes of the rats in aspergillus keratitis group infected for 12 h,white infiltrates and bloodshot in the eyes in 24 h group and the white infiltrates were further strengthened and almost covered the entire eyes in 48 h group. The order of inflammatory cytokines IL-1,TNF-α,IL-6,IL-10,NF-κB was:48 h group >24 h group >12 h group >control group(P<0.05),so did the expression levels of Dectin-1 and TLR4 protein and mRNA in various groups tested by Western blotting and RT-PCR methods.The expression levels of Dectin-1 and TLR4 were increased with the prolongation of time investigated by immunohistochemical staining. Conclusion Dectin-1 and TLR4 are secreted in excess after the eyes are infected with fumigatus,which promotes the expressions of related inflammatory cytokines and results in the occurrence of keratitis.

Influence of icaritin in proliferation and apoptosis of NB4 leukemia cells and its mechanism

Objective To investigate the influence of icariin (ICA) on the proliferation and apoptosis of acute promyelocytic leukemia NB4 cells, and to explore its mechanism. Methods The NB4 cells at logarithm growth phase were selected and randomly divided into blank control group,different doses (4,8,16,32,64 μmol·L-1) of ICA groups.Afer the NB4 cells were treated with ICA for different time (24,48,72 h),MTT assay was used to detect the inhibitory rates of NB4 cells,flow cytometry was used to analyze the apoptosis and the cell cycle of the NB4 cells,and the expression levels of apoptosis-associated proteins were detected by Western blotting method. Results Afer the cells were cultivated for different time(24,48,72 h),the inhibitory rates of the cells in different doses of ICA groups were higher than those in control group (P<0.05) and they were increased in a dose-and time-dependent manner (P<0.05).The apoptotic rates of NB4 cells in different doses of ICA group 48 h after treatment were (6.52±4.12)%,(10.07±2.36)%,(15.41±3.64)%,(25.18±1.24)% and (29.41±1.43)%,and they were higher than that in control group (2.39%±1.87%) (P<0.05).The flow cytometry results showed that after the cells were cultivated for 24 h in different doses of ICA groups,the number of NB4 cells at S phase was lower than those in control group (P<0.05),and the number of NB4 cells at G1 phase was increased (P<0.05). The expression levels of Bcl-2 in ICA groups were significantly lower than that in control group (P<0.05),while the expression levels of Bax were higher than that in control group (P<0.05). Conclusion ICA can induce the apoptosis of NB4 cells via inhibiting the expression level of Bcl-2 and up-regulating the expression level of Bax.

Expressions of OPN and CXCR4 in human epithelial ovarian cancer tissueand their clinical significances

Objective To analyze the expressions of osteopontin(OPN) and chemokine(CXC-subfamily)receptor 4(CXCR4) in different ovarian tissues,and to explore the role of OPN and CXCR4 in occurrence,development and metastasis of human ovarian cancer. Methods The expressions of OPN and CXCR4 in normal ovarian tissue(n=20),benign ovarian epithelial tumor(n=20) and epithelial ovarian cancers tissues(n=40) were detected by immunohistochemical SP method, and the expression rates of OPN and CXCR4 in epithelial ovarian cancer tissue with different clinical characteristics were analyzed. Results The positive expressions rate of OPN in epithelial ovarian cancer tissue was significantly higher than those in normal ovarian tissue and benign ovarian tumor tissue,and there were significant differences in the positive expression rates of OPN between different pathological stages (G1,G2,G3) of epithelial ovarian cancer (P<0.05).Meanwhile,the positive expression rate of OPN in Ⅲ-Ⅳ epithelial ovarian cancer tissue was significantly higher than that in Ⅰ-Ⅱ epithelial ovarian cancer tissue (P<0.05).However,there were no significant differences of the positive expression rates of OPN among different histological types of epithelial ovarian cancer tissue (P>0.05).There was no positive expression of CXCR4 protein in normal ovarian tissue and benign ovarian tumor tissues,but there was positive expression in epithelial ovarian cancer tissue.Besides,the expressions of CXCR4 protein were not significantly different among different pathological grades,different clinical stages and different histological types (P>0.05). Conclusion The OPN and CXCR4 expression levels are correlated with the degrees of malignancy in epithelial ovarian tumor,therefore the CXCR4 and OPN expression pathways may be the new targets for ovarian cancer therapy.

Objective To investigate the inhibitory effect of Rhodiola total flavonoids on transforming growth factor (TGF)-β1-induced proliferation of cardiac fibroblasts (CFB) in the rats,and to explore its mechanisms of improving myocardial fibrosis. Methods 5 μg·L-1 TGF-β1 was used to induce the proliferation of CFB to build the cell models of myocardial fibrosis.The cultured CFB were divided into control group,model group,and low,medium,high doses(25,50 and 100 mg·L-1) of Rhodiola total flavonoids groups.The cells were treated for 48 h,the vitalities of cells were detected by MTT, the levels of collagen proteinⅠ (ColⅠ) and collagen proteinⅢ (Col Ⅲ) in supernatant were measured by ELISA,and the activities of total superoxide dismutase (T-SOD) and glutathione peroxidase (GSH-Px) were detected, the levels of malondialdehyde (MDA) and glutathione (GSH) were also measured. Results The MTT results indicated that compared with control group,the cell vitality in model group was significantly increased (P<0.01). compared with model group, the cell vitalities of CFB in Rhodiola total flavonoids groups were significantly decreased (P<0.05).Compared with control group,the T-SOD activity in model group was decreased,while the MDA level was significantly increased (P<0.05); compared with model group,the T-SOD activities in 50 and 100 mg·L-1 Rhodiola total flavonoids groups were increased (P<0.01); the MDA levels in 25 and 50 mg·L-1 groups were decreased significantly (P<0.05).Compared with control group,the GSH-Px activity and GSH level in model group were significantly decreased (P<0.01). Compared with model group, the GSH-Px activities and GSH levels in Rhodiola total flavonoids groups were increased (P<0.05).Compared with control group, the ColⅠand Col Ⅲ levels in model group were significantly increased (P<0.01); compared with model group, the ColⅠand Col Ⅲ levels in Rhodiola total flavonoids groups were significantly decreased (P<0.05). Conclusion Rhodiola total flavonoids can inhibit the proliferation of rat CFB.The development of myocardial fibrosis may be inhibited by Rhodiola total flavonoids through anti-oxidative stress pathway.

Objective To construct the angiotensinogen (AGT)-targeting RNA interfering plasmids,and to explore the effect of AGT gene on the differentiation of preadipocytes 3T3-L1 in the mice. Methods The small hairpin RNA (AGT-shRNA) interference plasmids,with a scrambled Non-shRNA as control,were constructed on the basis of the activity of U6 promoter-driven expression vector psiRNA-U6.1/Neo and transfected into the mouse preadipocytes 3T3-L1.Two cell lines were generated from stable transfections.RT-PCR and SDS-PAGE were performed to monitor the mRNA and protein expressions of AGT gene, respectively.The effect of adipose AGT on the differentiation of 3T3-L1 was investigated through microscopic observation and profiling of adipocyte differentiation markers. Results Both AGT-shRNA and Non-shRNA interference vectors were successfully constructed; the expression of AGT mRNA was inhibited(P<0.05)and the expression of AGT protein was 43.86% of control.Both adipocyte differentiation markers,intracytoplasmic lipid levels and glycerol-3-phophate dehydrogenase (GPDH) activities were increased during the differentiation of preadipocytes 3T3-L1,but the data were lower than those in control group(P<0.05). Conclusion AGT gene silencing can significantly inhibit the differentiation of preadipocytes 3T3-L1,which demonstrates that there is important relationship between adipocyte metabolism and renin-angiotensin system (RAS) in adipose tissue.

Objective To study the inhibitory effect of fluorouracil combined with DDP on the growth of human osteosarcoma cell line MG-63, and to explore its influence on the expressions of transient receptor potential vanilloid 5 (TRPV5) and transient receptor potential vanilloid 6 (TRPV6) proteins. Methods The MG-63 cells were cultured by the density of 5×104 mL-1.Fluorouracil group, DDP group, fluorouracil+DDP group and control group containing 10% FBS were set up.The inhibitory rates of growth of MG-63 cells at different time were detected by CCK-8 assay.The apoptosis of MG-63 cells after treated with different drugs was determined by Hoechst staining Kit.The immunocytochemical staining was used to treatent to detect the expressions of TRPV5 and TRPV6 before and after treatment. Results Fluorouracil and DDP both inhibited the growth of MG-63 cells in a time-and dose-dependent manner.There were a lot of black particles in the MG-63 cells and the cells were smaller, aging or death when they were exposed to fluorouracil or DDP.Compared with 24 h group, the inhibitory rates of proliferation of MG-63 cells after treated with the sigle drug of fluorouracil or DDP for 48 and 72 h were increased significantly (P<0.05).Compared with control group,the apoptotic rates of MG-63 cells in fluorouracil group and DDP group 24,48,and 72 h after treatment were increased (P<0.01) in a time-dependent manner.The expression levels of TRPV5 and TRPV6 in MG-63 cells 72 h after treatment of fluorouracil and DDP were decreased significantly compared with before treatment(P<0.05). Conclusion Fluorouracil, DDP and fluorouracil combined with DDP could significantly inhibit the proliferation of MG-63 cells, induce the apoptosis, and decrease the expression levels of TRPV5 and TRPV6.

Influence of different surface treatments in bond strength of brackets to porcelain

Objective To observe the porcelain treated with Nd∶YAG laser irradiation and hydrofluoric acid(HF), and to explore the influence of integrated treatment of Nd∶YAG laser irradiation and HF etching in the bond strength of brackets to porcelain. Methods 48 metal ceramic prostheses were randomly divided into untreated control group, HF group, grooved treatment group, 0.75W laser group, 1.05W laser group, 1.45W laser group. All samples were bonded to the brackets. After temperature cycling test, the shear bond strength (SBS) and tensile bond strength (TBS)were measured. Results There were significant differences in SBS and TBS between various surface treatment groups and untreated control group (P<0.01). The SBS and TBS of brackets bonded in HF group was significantly higher than those in 1.05W laser and 1.45W laser groups (P<0.05).The SBS and TBS in 1.05W laser and 1.45W laser groups were higher than those in HF group(P<0.05). The SBS and TBS in 1.05W laser group were higher than those in other groups(P<0.05). SBS showed positive correlation with TBS(r=0.426, P=0.000). Conclusion The use of Nd∶YAG laser irradiation with the energy parameter of 1.05W and HF could increase the bonding with formation of composite resin, and the more SBS, the more TBS.

Detection of human papillomavirus DNA genotypes in different populations and its clinical significance

Objective To analyze the detection results and clinical significance of human papillomavirus(HPV) DNA genotypes in the patients with condyloma acuminate(CA) and high risk(HR) population of CA and normal population of physical examination(PE). Methods 527 cases of CA patients (CA group), 208 cases of HR (HR group) and 197 cases of normal PE (PE group) in the outpatient were selected. The human HPV DNA genotypes in 932 samples of different population were examined with HybriMax; the positive detection rates, the preponderance types of HPV and the multiple genotype infection in three groups were analyzed. Results The HPV positive rate in CA group was 98.29% and the preponderance types were HPV 6,11,33,and 52 genotypes. The HPV positive rate in HR group was 26.44% and the preponderance types were HPV 6,11,33,and 66 genotypes. The HPV positive rate in PE group was 5.58% and the preponderance types were HPV 6 and 11 genotypes. The difference in the positive detection rates of HPV between HR and PE groups had statistical significance(P<0.05).The positive rates of multiple genotype infection in CA group and HR group were 36.43% and 5.77%, respectively, and there was significant difference (P<0.01);the positive rate of double infection was the highest (18.41%), followed by the positive rate of triple heavy infection (11.20%) and the positive rate of septuple heavy infection (0.38%). Conclusion The HPV 6 and HPV 11 of low-risk HPV genotypes are the most common genotypes in different population. The HR group and PE group are all asymptomatic but the positive detection rate of HPV in HR group is higher than that in PE group. The positive rate of HPV multiple genotypes infection in CA group is higher than that in HR group, and the double infection or triple heavy infection are the most common infection.

Objective To explore the effectiveness and safety of totally laparoscopic distal gastrectomy (TLDG) and laparoscopically assisted distal gastrectomy (LADG) for gastric cancer. Methods The comparative studies of TLDG and LADG published between 2008 and 2014 were searched from PubMed,EMBASE,Chinese Biomedical Literature Database (CBM),China National Knowledge Infrastructure (CNKI).After screening for inclusion,data extraction,and quality assessment,RevMan 5.3 software was used for Meta-analysis. Results Ten studies of 2 212 patients were included in the Meta-analysis,among whom 930 cases underwent TLDG and 1 282 cases underwent LADG.The results of Meta-analysis indicated that compared with LADG,TLDG had the advantages of less blood loss(WMD=-20.70,95%CI:-30.81--10.59,P<0.01),less usage of analgesic(WMD=-0.38,95%CI:-0.74--0.02,P=0.04),more retrieved lymph nodes(WMD=2.98,95%CI:0.71-5.26,P=0.01).However,the Meta-analysis showed no statistically significant differences in the operation time,postoperative time-to-first flatus and oral intake,postoperative hospital stay,length of proximal resection margin,C reaction protein(CRP) level at postoperative day 1,incidence of overall complications and anastomosis-related complications. Conclusion TLDG is safe and effective with less blood loss,less pain than those of LADG.Moreover,it has comparable results to conventional LADG,with no increase of postoperative complications.

Relationship between serum levels of galectin-3 and prognosis of patients with chronic heart failure:A Meta-analysis

Objective To evaluate the relationship between the newly recominended chronic heart failue(CHF) marker-serum galection-3(Gal-3) level and the prognosis of CHF by Meta-analysis. Methods The Chinese and English literatures about the relationship between Gal-3 and CHF prognosis were searched in PubMed,EMbase,Chinese Biomedical Literature Databases.All literatures were assessed by the inclusion and exclusion criteria.Hazard ratio (HR) and 95% confidence interval (CI) in each literature were extracted for Meta-analysis using STATA 12.1 software.The stability and reliability were evaluated by sensitivity analysis and publication bias analysis. Results Ten literatures complying with the requirements were included.The results of heterogeneity test were as follows: ① When the influence of other factors were not excluded, heterogeneity test showed that P=0.000, I2=89.2%, heterogeneity existed in different studies, so statistics were combined using a random effects model. The combined HR of univariate analysis was 1.36 and 95% CI was 1.23-1.50. 2. When the influence of other factors were excluded, heterogeneity test showed that P<0.001,I2=68.4%, heterogeneity existed in different studies, so statistics were combined using a random effects model. The combined HR of multivariate analysis was 1.17 and 95% CI was 1.07-1.26. 3. Sensitivity analysis showed that when the study with the largest samples was excluded, the combined HR of univariate analysis was 1.54 and 95% CI was 1.32-1.77; the combined HR of multivariate analysis was 1.26 and 95% CI was 1.09-1.42. When the study with the smallest samples was excluded, the combined HR of univariate analysis was 1.34 and 95% CI was 1.20-1.47; the combined HR of multivariate analysis was 1.15 and 95% CI was 1.06-1.24. Conclusion The serum level of Gal-3 is increased significantly in the patients with CHF and it may be an independent risk factor for poor prognosis of the CHF patients.

Objective To investigate the association between rs2281951 and rs3798753 in ELOVL fatty acid elongase2 (ELOVL2 gene) and the docosahexenoic acid(DHA)level in breast milk,and to clarify the influence of the polymorphisms of ELOVL2 gene in the DHA level of breast milk. Methods 209 healthy maternals were selected and signed the consent form and completed the 3-day 24-hour dietary recall questionaire on one day during the 22nd-the 25th day after partum, and 20 mL breast milk was collected.The DHA level in breast milk was detected with gas chromatography.The milk DNA was extracted and two single nucleotide polymorphisms(SNPs)of ELOVL2 gene were detected by Sequenom Mass Array System.UNPHASED 3.012 genetics software was adopted to analyze the quantitative trait of haplotype and the DHA level in breast milk. Results The distribution of genotypic frequencies of rs2281591 and rs3798713 sites in ELOVL2 gene was consistent with Hardy-Weinberg equilibrium (P>0.05).The dietary fatty acid intakes and the milk DHA levels of maternals carrying different genotypes had no statistically significant differences (P>0.05).The DHA levels in breast milk of maternals carring different rs3798713 (CG)-rs2281591 (AG) haplotypes had no statistically significant difference (χ2= 3.422,df=5,P=0.635). Conclusion Rs3798713 and rs2281951 and constructed haplotypes in ELOVL2 gene are not related to the DHA levels in breast milk.

Detection of serum folate, homocysteine and vitamin B12 in patients with non-smallcell lung cancer and its clinical significance

Objective To detect the serum levels of folate, homocysteine (Hcy) and vitamin B12 in the patients with non-small cell lung cancer(NSCLC), and to clarify the clinical significance of folate,Hcy and vitamin B12 in the diagnosis and prognosis assessment of NSCLC. Methods 35 patients with NSCLC were chosen as NSCLC group,and 30 healthy people were selected as control group, excluding hypertension, anemia, family disease history and other related factors. The expression levels of serum folate,Hcy and vitamin B12 were examined by circulating enzymatic method and electrochemical luminescence method.The correlations between the levels of serum folate,Hcy and vitamin B12 of the objects in two groups were analyzed by Pearson test. Results The serum Hcy level of the patients in NSCLC group was significantly higher than that in control group(P<0.01).The serum folate level of the patients in NSCLC group was significantly lower than that in control group(P<0.01),while there was no significant difference in vitamin B12 levels between two groups(P>0.05).The serum Hcy level was negatively correlated with folate level(r=-0.505,P=0.002), but was not correlated with vitamin B12(r =-0.084,P=0.633). The serum folate level was not correlated with vitamin B12(r=-0.039,P=0.826). Conclusion The serum Hcy level of NSCLC patient is significantly increased and it has diagnostic and prognostic values in the NSCLC patients.

Analysis on related factors of axillary lymph node metastasis in patients with cT1-2N0 breast cancer

Objective To investigate the relationships between axillary lymph node metastasis and clinicopathologic features in the patients with cT1-2N0 breast cancer and clarify the law of axillary lymph node metastasis,and to find the risk factor,and provide the theoretical basis for individuation therapy. Methods 687 patients with cT1-2N0 breast cancer were divided into negative group and positive group according to the pathological results of axillary lymph node, and the clinicopathologic features were layered.The risk factors of axillary lymph node metastasis were screened out by Chi-square test and Logistic regression analysis. Results In 687 cases of cT1-2N0 breast cancer,156 (22.7%) cases were observed with axillary lymph node metastasis. The age,cT stage,pT stage,pathological type,vascular invasion,perineural invasion estrogen receptor(ER), progesterone receptor(PR),and molecular subtyping were the factors that influenced axillary lymph node metastasis in univariate analyses.The age<35 years,cT2,invasive ductal carcinoma,vascular invasion positive and Luminal subtyping were the independent risk factors of axillary lymph nodes metastasis in multivariate analyses(r=3.440,P=0.010;r=1.770,P=0.007;r=3.397,P=0.001;r=7.434,P=0.000;r=2.212,P=0.015). Conclusion In the patients with cT1-2N0 breast cancer,the age, cT,pathological type, vascular invasion and molecular subtyping are important predictors of axillary lymph node metastasis and vascular invasion was the most important predictor.The preoperative comprehensive analysis and evaluation of clinical data and preoperative pathological results obtained will help to select the right surgical operation.

Influence of administration of dexmedetomidine before uterectomy in stress and immunologic function in patients

Objective To explore the effect of dexmedetomidine on the stress and immunologic function in the patients undergoing uterectomy,and to provide an evidence for the medication in clinical anesthesia. Methods A total of 60 hysteromyoma patients who needed uterectomy were selected. All the patients were in ASAⅠ-Ⅱlevel.The patients were divided into observation group (n=30) and control group (n=30) according to administration of different types of drugs.Before anesthesia,the patients in observation group were given dexmedetomidine (l μg·kg-1) with a concentration of 4 g·L-1 for 10 min,while the patients in control group were given equal normal saline.The induction was performed 10 min after infusion pump.Sevoflurane and fentanyl were used for maintaining during the anesthesia.The blood samples of the patients in two groups before entering the operation room (T0),2h after operation (T1),and extubation after operation (T2) were collected.The serum cortisol (COR) and interleukin-2 (IL-2) concentrations,plasma T lymphocyte subpopulations and NK levels were detected. Results The differences of COR and IL-2 concentrations at T0 between two groups were not statistically significant (P>0.05).The COR concentrations at T2 in two groups were significantly higher than those at T0 (P<0.05).The COR concentration at T2 in observation group was significantly lower than that in control group (P<0.05).The serum IL-2 concentration was gradually decreased with the prolongation of time observation in two groups.The serum IL-2 concentration in observation group was significantly higher than that in control group (P<0.05).The differences of CD3+,CD4+,CD8+,CD4+/ CD8+,and NK levels at T0 between two groups were not statistically significant (P>0.05).The differences of CD8+ level in each time point between two groups were not statistically significant (P>0.05).The CD3+,CD4+,CD4+/ CD8+,and NK levels at T1 and T2 in control group were significantly lower than those at T0 (P<0.05).The CD3+,CD4+,CD4+/ CD8+,and NK levels at T2 in observation group were significantly higher than those in control group (P<0.05). Conclusion Dexmedetomidine assisting anesthesia can alleviate the postoperative stress reaction,and reduce the effect of anesthesia and operation wound on the cellular immune function.It is beneficial for the postoperative rehabilitation.

Relationships between vertical facial pattern and natural head position,cervical posture in patients with skeletal class Ⅰ relationship

Objective To investigate the natural head position and cervical posture in the patients with different vertical facial patterns with skeletal class Ⅰ relationship, and to clarify the relationships between vertical facial patterns and natural head position, cervical posture in the patients with skeletal class Ⅰ relationship. Methods 94 patients aged 8-15 years old with skeletal class Ⅰ relationship were selected as the subjects. The subjects were classified into high angle group, average angle group, and low angle group according to their mandibular plane angles. The cephalometric radiographs in natural head position were taken, and the variables representing craniofacial morphology, head position, and craniocervical posture were measured. The intergroup differences in variables were compared. Results The inclinations of mandible to the true vertical and cervical column (ML/VER,ML/OPT,ML/CVT) were smallest in high angle group, and they were largest in low angle group (P<0.01); other variables representing head posture and craniocervical posture (NSL/VER,FH/VER,NSL/OPT,NL/OPT, NSL/CVT,FH/CVT) were largest in high angle group, and they were smallest in low angle group (P<0.05). The inclinations of ramus to cervical column (RL/OPT,RL/CVT) were largest in high angle group and they were smallest in low angle group (P<0.01). There were no significant differences in the inclinations of cervical column and cervical lordosis (OPT/HOR,CVT/HOR,OPT/CVT) among three groups (P>0.05). Conclusion The significant differences exist in natural head position and cervical posture among the patients with skeletal class Ⅰ relationship with different vertical facial patterns (high angle,average angle,low angle); the subjects with high angle show extended head position and cervical posture, while the subjects with low angle exhibit the opposite tendency.

Clinical effect and safety evaluation of microsurgery associated with Gamma Knife radiosurgery for large meningiomas in sellar region

Objective To investigate the therapeutic effect and safety of microneurosurgery associated with Gamma Knife radiosurgery on the large meningiomas in sellar region,and to clarify its clinical curative effect. Methods The clinical data of 34 patients with large meningiomas in sellar region underwent microsurgery were retrospectively analyzed.All of them underwent microsurgery,and then treated with Gamma Knife radiosurgery in one month after operation if there were residual tumors.The tumor removal of situation,complication,rate of symptom remission,and recurrence rate were analyzed. Results Among the 34 patients,total resection(7 cases of Simpson grade Ⅰ and 12 cases of Simpson grade Ⅱ) was achieved in 19 cases(55.9%),subtotal resection (Simpson grade Ⅲ ) in 14 cases (41.2%),and partial resection (Simpson grade Ⅳ) was achieved in 1 case (2.9%).The major complications were cranial nerve injuries(such as oculomotor nerve,trochlear nerve and abducens nerve,n=6),the contralateral limb paresis(n=2),postoperative bleeding(n=1),CSF leak with infection(n=3),and secondary epilepsy(n=3);no death occurred postoperatively.All these patients were followed up for about 3 to 48 months.Postoperative headache disappeared in 24 cases (80%),and 6 cases were alleviated(20%);postoperative vision improved in 12 cases (80%),remained unchanged in 2 cases(13.3%) and deteriorated in 1 case(6.7%);postoperative olfactory function improved in 5 cases(62.5%) and 3 cases unchanged(37.5%);5 cases recoverd from the ocular motility disorder (71.4%),and 2 cases unchanged(28.6%);7 cases recoverd from thehemiplegia(100%).One of the total resection cases(5.2%) and two of the non-total resection(13.3%) suffered from tumor recurrence.All of the non-total resection cases were treated with Gamma Knife radiosurgery. Conclusion The clinical effect of microneurosurgery associated with Gamma Knife radiosurgery in treatment of large meningiomas in sellar region is satisfactory.

Objective To observe the clinical effect of rosuvastatin inthe treatment of lower extremity atherosclerotic disease in the elderly,and to provide clinical basis of the application of statins in lower extremity atherosclerotic disease in the elderly. Methods 90 patients diagnosed as lower extremity atherosclerotic disease (aged 60 years or over)were randomly divided into simvastatin group and rosuvastatin group (n=45),and they were treated with simvastatin and rosuvastatin, respectively, based on the regular treatment.The total cholesterol(TC),low density lipoprotein-cholesterol(LDL-C),and high sensitivity C-reactive protein (hs-CRP) of the patients in two groups were detected at the begining of treatment and the ending of 32 weeks;at the same time,the lower limb artery intima thickness,strength of hardened,plaque,and artery stenosis degree scores were compared. Results 32 weeks after treatment, the levels of TC,LDL-C and hs-CRP of the patients in two groups were decreased statistically compared with before treatment (P<0.05);the levels of TC,LDC-C,and hs-CRP in rosuvastatin group were lower than those in simvastatin group(P<0.05).32 weeks after treatment in two groups,compared with before treatment,the lining thickness and plaque,stenosis scores were decreased (P<0.05); the lining thickness,plaque and stenosis scores in rosuvastatin group were lower than those in simvastatin group (P<0.05);the gardening strength grades had no statistical difference between before and after treatment. Conclusion Statins is lipid-lowering and resistant to lower limb arteriosclerosis,and rosuvastatin is better than simvastatin.

Application of neural electrophysiological monitoring in facial nerve microvascular decompression

Objective To expore the application of intraoperative neural electrophysiological monitoring in microvascular decompression (MVD) of idiopathic facial spasm(HFS),and to clarify the effect of MVD in increasing the operative efficacy of MVD and decreasing the postoperative complications. Methods From December 2010 to December 2014, 163 patients with HFS received MVD were selected;from December 2010 to December 2010,73 patients with facial spasm without electrophysiological monitoring in the operation were used as control group; from January 2013 to December 2014,90 patients with facial spasm with BAEP/LSR/FN MEP monitoring in the operation were used as monitoring group.The postoperative efficiency of MVD and the occurrence of hearing loss,dizziness,facial paralysis and other complications after operation were compared between two groups. Results The immediate efficiency in monitoring group was 52.55% (47 cases),minor facial paralysis 1.11% (1 case),hearing loss and dizziness 5.56% (5 cases).The postoperative follow-up time was 6 to 12 months,an average of 9.6 months;the facial paralysis,hearing loss,and dizziness were significantly improved and the operation efficiency was 65.56% (59 cases).The Immediate efficiency in control group was 30.14%(22 cases),minor facial paralysis 13.69% (10 cases),hearing loss and dizziness 23.29% (17 cases); the postoperative follow-up time was 6 to 12 months,an average of 9.6 months; the facial paralysis,hearing loss,and dizziness were significantly improved,and the operation efficiency was 64.38% (47 cases).The immediate surgery had statistically significant difference between two groups(P<0.05),and monitoring group was better than control group; the long-term curative effect had no significant difference between two groups (P>0.05).The incidence of facial paralysis,hearing loss and the incidence of complications such as dizziness had statistically significant differences between two groups (P<0.05),and monitoring group was better than control group. Conclusion LSR monitoring can improve the short-term curative efficiency of facial nerve MVD,but the significance for the long-term curative efficiency is not obvious;BAEP,LSR and FN MEP monitoring have great significance in identification of responsibility vessel,judgement of decompression effect and surface and auditory nerves protection.

Application of bilevel positive airway pressure in treatment of respiratory distress syndrome in preterm infants

Objective To explore the influence of intubation-surfactant-extubation (InSurE) therapy combined with bilevel positive airway pressure (BiPAP) in the use time of mechanical ventilation,and to clarify the value of BiPAP in the treatment of respiratory distress syndrome in the preterm infants. Methods Toral 95 preterm infants with respiratory distress syndrome were treated with InSurE therapy during January 2011 to October 2014.Among them,the preterm infants before January 2013 were selected as control group who were treated with InSurE and nasal continuous positive airway pressure (nCPAP).After January 2013,60 preterm infants were treated with BiPAP,as BiPAP group.The rates of InSurE failure,the need for mechanical ventilation (MV) on the 7th day after InSurE failure,total non-invasive ventilation time,total mechanical ventilation time,atmospheric oxygen therapy time and incidence of clinical complications were compared between two groups. Results 1 There were no significant differences in the clinical data of the preterm infants between two groups,such as gender and age.2 Although there was no significant difference in the failure rate of InSurE,but the rate of repeated mechanical ventilation during 1 week in BiPAP group was lower than that in control group (P<0.01).3 The Rank sum test result showed that the total time of non invasive ventilation in BiPAP group was longer than that in control group (P<0.01).The total time of invasive mechanical ventilation and oxygen therapy in BiPAP group was lower than that in control group (P<0.05).4 The incidence of retinopathy of prematurity (ROP) and bronchopulmonary dysplasia (BPD) in BiPAP group was lower than that in control group. Conclusion BiPAP can significantly reduce the use of invasive mechanical ventilation after the failure of InSurE,thereby decreases the oxygen toxicity and barotrauma hazards.

Application of 18FDG PET/CT in diagnosis of ectopic ACTH syndrome induced by lung cancer

Objective To investigate the value of 18F-fluorodeoxyglucose(18FDG) positron emission tomography and computer tomography(PET/CT) in the diagnosis and localization of ectopic ACTH syndrome(EAS) induced by lung cancer, and to increase the accuracy rate of diagnosis of EAS. Methods 8 patients were histopathologically proven to be lung cancer. 18FDG PET/CT wholebody and brain scan were performed; magnetic resonance imaging(MRI), 99mTc-octreotide(OCT) imaging and inferior petrosal sinus sampling(IPSS) were done in all patients. On 18FDG PET/CT images, the SUVmax values of lesions and the SUVmax ratio of lesion tissue to normal lung tissue were calculated. Results ① 8 patients with EAS included 2 cases of carciniod, 4 cases of atypical carciniod, and 2 cases of small cell lung cancer. The ACTH levels in inferior petrosal sinus and peripheral veins of 8 patients had no difference.②The chest CT showed there were pulmonary nodules in 6 cases (75%), among them 2 cases were atypical lesions. 5 cases of pituitary adenomas were found by MRI(25%), and OCT imaging was positive in 2 cases (25%).③ The 18FDG PET/CT showed nodular, mass like or patch samples with high metabolic activity in 8 cases (100%) of pulmonary lesions. The SUVmax of tumor was 0.72 to 14.05 (3.67±4.68), and the ratio of SUVmax of lesion tissue to normal lung tissue was 1.69 to 27.54 (8.14± 9.63). 18FDG hypermetabolism was found in 8 cases of bilateral adrenal hyperplasia, while 4 cases showed pituitary tumor. Conclusion Lung cancer is a common source of EAS, especially carcinoid. 18FDG PET may show lower glucose metabolism. 18FDG PET/CT should be used as an assisted method to diagnose EAS.

Objective To measure the morphological parameters of cervical endplate of Chinese by using computed tomography (CT) scans, and to provide an accurate morphometric basis for designing and developing the cervical disc prostheses. Methods 80 healthy subjects were scanned by CT. The parameters of each cervical vertebra from C3 to C7 were measured by CT scans including upper anteroposterior length (APLu), upper center mediolateral length (CMLu), lower anteroposterior length (APLl) and lower center mediolateral length (CMLl). These parameters were compared between genders and among different vertebral levels. Results The values of APLu, CMLu, APLl and CMLl were increased with the decreasing of cervical segment (PAPLu=0.023,PCMLu=0.007,PAPLl=0.035, PCMLl<0.001). There was statistically significant difference in morphological parameters between genders (P<0.05).Compared with the reported data of other different populations,the statistically significant difference in morphological parameters also existed in the study. Conclusion The study provides an accurate morphological basis for designing the suitable artificial cervical disc for Chinese population.

Objective To investigate the changes of waist muscle density and area in the subjects with spiral CT scan examination,and through the quantitative analysis of different age and gender to get the degree of lumbar muscle degeneration. Methods 240 cases of lumbar CT scan reconstruction images (including the patients suffering from low back pain)were collected.According to the age, 20-29 years old group,30-39 years old group,40-49 years old group, 50-59 years old group,60-69 years old group,and 70-79 years old group were set up. The lumbar transverse psoas muscle, lumbar side muscle, erector spinae cross-sectional areas(CSA) and mean CT values were measured at different levels. Scanning equipment: Toshiba Activion 16-slice spiral CT scanners, acquisition range from the waist 1 to S1. Results ①Compared between the measurement results of the same age groups(except 70-79 years old group),the average CSA of the male at the same level waist was higher than the female(P<0.05).②Compared between the same level groups of 20-29 years,30-39 years and 40-49 years,there were no significant differences(P>0.05). Compared between the same level groups of 50-59 years,60-69 years and 70-79 years,there was no significant difference(P>0.05). ③Compared the CSA of man's erector spinae muscle between 40-49 years group and 50-59 years group, there were significant differences(P<0.05). ④Compared the same level CSA of female between 20-29 years,30-39 years,40-49 years,50-59 years,60-69 years,70-79 years groups,and the adjacent groups,there were no significant differences(P>0.05). ⑤The same level average density of erector spinae muscle was lowest compared with the psoas major muscle and quadratus lumborum muscle, and falling faster, the psoas major muscle was relatively higher. The same level average density of female's erector spinae muscle was lower than man's,specially in 70-79 years group. Conclusion Multi-slice spiral CT can be used to diagnose the lumbar degenerative conditions.

Objective To investigate the correlation between the quantitative results, the semi-quantitative scores from ultrasound elastography and the distribution of myofibroblasts(MFS) in breast tumor,and to analyze the value of quantitative results and the semi-quantitative score from ultrasound elastography in the diagnosis of breast tumor. Methods Thirty eight patients with breast lesions underwent ultrasound elasticity examinations, tissue dispersion quantitative analysis technique was used to assess the 11 characteristic quantities and the corresponding strain ratios in all lesions,and the score of ultrasonic elastography was evaluated.38 cases were divided into benign and malignant group according to pathological diagnosis results.The expression levels of CD34 and α-SMA protein in breast tissue were examined by immunohistochemistry.The distribution patterns of MFS in breast tumor were analyzed.The correlation between the quantitative results, the semi-quantitative scores from ultrasound elastography and the distribution of MFS in breast tumor was studied. Results The expression level of CD34 in malignant group was significantly higher than that in benign group,while the expression level of α-SMA was significantly higher than that in benign group;the differences in the expression levels of CD34 and α-SMA between benign and malignant breast tumor patients were statistically significant(P<0.01). The masses observed in malignant group by elastography were shown in blue,while most of the masses in benign group were shown in green.There were statistically significant differences between two groups in elastography scores(P<0.05).There was a anegative correlation between the CD34 expression in tumor tissue and the score of ultrasound elastography (r=-0.423 7,P=0.027 3).There was a negative correlation between the CD34 expression and the score of ultrasound elastography(r=-0.423 7,P=0.027 3),while the positive correlation was found between the α-SMA expression and the score of ultrasound elastography(r=0.397 0,P=0.014 2).The CD34 expression was significantly correlated with the average relative strain value, entropy, area ratio of low-strain region, kurtosis, skewness and inverse difference moment (P<0.05).Among the 11 characteristics, CD34 was positively correlated with the average relative strain value(r=0.385 6,P=0.016 8) and entropy(r=0.380 5,P=0.018 5);CD34 was negatively correlated with area ratio of low-strain region(r=-0.351 7,P=0.030 4), kurtosis(r=-0.427 7,P=0.007 4), skewness(r=-0.394 6,P=0.014 2),inverse difference moment(r=-0.344 3,P=0.034 3), angular second moment(r=-0.484 9,P=0.002 0) and strain ration(r=-0.379 0,P=0.047 5);CD34 was not correlated with standard deviation, complexity and contrast(P>0.05). The α-SMA espression was positively correlated with kurtosis(r=0.356 9,P=0.027 8), skewness(r=0.323 0,P=0.047 9), area ratio of low-strain region(r=0.382 0,P=0.021 6)and strain ratio(r=0.403 3,P=0.012 0). Conclusion The features and its scores of ultrasound elastography are correlated with the distribution of MFS in breast tumor, suggesting that the ultrasound elastography is very informative and helpful in the diagnosis of breast tumor.

Distribution characteristics of toxic heavy metal pollution in cosmetics of beauty and hairdressing salons and their health risk assessment

Objective To explore the method of health risk assessment for heavy metals in cosmetics and to understand the situation of the toxic heavy metals pollution in cosmetics of beauty and hairdressing salons,and to clarify the pollution causes and its potential health risks. Methods 27 beauty and hairdressing salons were randomly sampled from a city in Gansu province (11 beauty salons and 16 hairdressing salons).The 18 samples of whitening cream,the 13 samples of lip gloss,the 14 samples of hair agent and the 16 samples of shampoo were selected from the 27 beauty and hairdressing salons,the total sample size was 61.The atomic fluorescence spectrometer and atomic absorption spectrometer methods were used to detect the contents of As,Pb,Hg and Cd in cosmetics,and the pollution levels were compared between the current standard and new standard.The health risk assessment models were employed to assess the health risk of those unqualified cosmetics. Results The average contents of As,Pb,Hg and Cd in 61 samples were all below the current allowable limit,and the average contents of Pb in the samples of whitening cream,the hair agent and the shampoo exceeded the new allowable limit.The pollution levels were slight-pollution for most of unqualified cosmetics.The results of health risk assessment showed that all of the cosmetics were security. Conclusion On the whole,the qualities of cosmetics that are used in beauty and hairdressing salon are qualified,but they still don't match the new standard well,and there are still a few of cosmetics unqualified.

Analysis on association between emotional quotient, personality trait and interpersonal trustamong students in higher university

Objective To explore the association between the students' emotional quotient(EQ),personality trait and interpersonal trust,and to clarify the potential function of interpersonal trust of higher students. Methods Interpersonal Trust Scale,Emotional Quotient Table and Eysenck Personality Questionnaire(EPQ) were used to investigate 364 students from Guangxi University and Guangxi Teachers Education University.SPSS 18.0 software was used for one-way ANOVA variance analysis,Pearson product-moment correlation analysis and stepwise Multiple Linear regression analysis. Results The differences in interpersonal trust,EQ and personality trait of EPQ had statistical significance between males and females,and the males' scores were higher than those of females(P<0.05,P<0.01);the differencs in Lie of EPQ between nations had statistical significance and the scores of Dong,Hui and Tujia ethnic minority were higher than those of other nationalities(P<0.01);the difference in personality trait of EPQ between educational background had statistical significances,and the undergraduates' scores were higher than those of master students(P<0.05).EQ was positively correlated with EPQ-E and EPQ-L (r=0.274,P<0.01;r=0.145,P<0.01),and was negatively correlated with EPQ-N and EPQ-P(r=-0.422,P<0.01;r=-0.202,P<0.01);interpersonal trust was negatively correlated with EQ and EPQ-E(r=-0.273,P<0.01;r=-0.169,P<0.01);special trust was negatively correlated with EQ and EPQ-E(r=-0.252,P<0.01;r=-0.151,P<0.01);interpersonal trust and general trust were negatively correlated with EPQ-P(r=0.313,P<0.01;r=0.338,P<0.01),and they were positively correlated with EPQ-L(r=-0.201,P<0.01;r=-0.217,P<0.01). Conclusion The students' EQ,personality trait are related to interpersonal trust of the students in Guangxi universities,and they have important prediction effects.Improving students' EQ and interpersonal trust plays a positive role,whereas,the possible negative effects accompanying should never be neglected.
