Anti-apoptosis effect of polycystin-1 gene on human cervical cancer HeLa cells

Abstract: Objective:To investigate the effect of polycystin-1(PC1)on the apoptosis of human cervical cancer HeLa cells,and to provide the basis for further clarifying the effect of PC1 on apoptosis. Methods: The recombinant plasmid pEGPF-PC1-5TMC of PC1 gene and blank control plasmid pEGFP were respectively used to transfect the HeLa cells,which was verified by Western blotting method.The proliferation activities of cells in transfection group (transfected with recombinant PC1 plasmid),blank control group(transfected with blank control plasmid pEGFP) and normal control group(normal HeLa cells)were respectively detected by MTT at 48 and 72 h after transfection.The apoptotic rates of the cells in transfection group and blank control group were detected by TUNEL method. Results: Compared with normal control group,the proliferation ability of HeLa cells in transfection group had no significant difference at 48 and 72 h after transfection(P>0.05);compared with blank control group,the proliferation ability of HeLa cells in transfection group was increased significantly (P<0.05) at 48 and 72 h after transfection.The apoptotic rate of HeLa cells in transfection group was obviously lower than that in blank control group (P<0.01). Conclusion: PC1 gene has anti-apoptosis effect on the human cervical cancer HeLa cells.
