Analysis of association between PTEN gene single nucleotide polymorphism |and stomach neoplasms

Abstract:

Objective To investigate the genetic assaciation between tow single nucleotide polymorphisms (SNPs) (rs2735343 and rs701848) of PTEN anti-oncogene and gastric carcinoma in Northern Han Chinese and to elucidate whether the two SNPs are susceptibility loci for gastric carcinoma in Northern Han Chinese.Methods The PCR-based restriction fragment length polymorphism (PCR-RFLP) analysis was used to detect these two SNPs present in the PTEN gene among 58 sporic gastric carcinoma patients (case group) and 104 normal individuals (control group).The association between the polymorphisms and gastric carcinoma was analyzed with SPSS 10.0.  Results The fragment of rs2735343 was 272 bp,and three genotypes (GG,GC,CC) were found after digestion;The fragment of rs701848 was 394 bp,and three genotypes (TT,TC,CC) were also found after digestion;The genotypic frequencies of rs2735343 and rs701848 were not deviated from Hardy-Weinberg equilibrium either in case group (P=0.062;P=0.055) or control group (P=0.246;P=0.692). The allelic and genotypic frequencies of rs2735343 and rs701848 had no remarkable difference between case and control groups (P>0.05).Conclusion Neither rs2735343 nor rs701848 of the PTEN gene is not associated with gastric carcinoma,they may not be susceptibility loci for gastric carcinoma in Northern Han Chinese.
