Expression of HIP1 in injuried Hela cells induced by vitamin K3 and significance

Abstract:

Abstract:Objective To observe the expression of Humtingtin interacting protein 1(HIP1) in the injury of Hela cells induced by vitamin K3(VK3).Methods Hela cells in log phase were randomly divided into control group,VK3 group,VK3+NAC group and NAC group,respectively.The survival rate of Hela cells was determined by MTT.The expressions of HIP1 and NF-κB protein  in Hela cells were detected by Western blotting.The expression of Cyclin D1 mRNA  was tested by RT-PCR assay.Results Compared with control group,the survival rate of Hela cells in VK3 group decreased(P<0.05);the  expressions of HIP1 and NF-κB protein and Cyclin D1 mRNA  were all decreased in VK3 group (P<0.05).Compared with VK3 group,the survival rate of Hela cells in  VK3 + NAC group was increased(P<0.05);the expressions of HIP1 and NF-κB protein  and Cyclin D1 mRNA  were increased(P<0.05).Conclusion VK3 has an effect of inhibiting the cell proliferation which may be related with down-regulation of HIP1 protein expression.
