Effects of purified product of trihydroxybenzoic acid and its compound on cell cycle progression and apoptosis of Jurkat cells in vitro

Abstract:

Abstract:Objective To study the effects of purified product of trihydroxybenzoic acid (TBA) from water caltrop and its compound at different concentrations on cell cycle progression and apoptosis of Jurkat cells in vitro and explore their possible mechanism of inhibiting tumor growth.  Methods The Jurkat cells at logarithmic growth phase were treated with purified product of TBA and its compound in vitro. Each TBA was divided into 5 groups (0,3.125,6.250,12.500 and 25.000 mg/L). The changes of cell cycle progression and apoptotic rate of Jurkat cells 30 h after treated with TBA were measured with flow cytometry.  Results The apoptotic rates of the Jurkat cells treated with purified product TBA with doses of 3.125 and 25.000 mg/L were increased significantly as compared with those in  control group(P<0.05 or P<0.01);the apoptotic rates of Jurkat cells treated with TAB compound with different doses were increased with the increase of doses (P<0.05 or P<0.01). The percentages of G1 phase cells treated with purified product TBA with different doses were increased significantly with the increase of doses as compared with those in  control group(P<0.01),the percentages of S phase cells were decreased significantly in 6.250 and  25.000 mg/L groups(P<0.05 or P<0.01),and the percentages of G2 phase cells did not change significantly. The percentages of G1 phase cells treated with TAB compound with 6.250 and  25.000 mg/Lwere significantly higher than those in  control group(P<0.05 or P<0.01),the percentages of S phase cells were decreased significantly (P<0.05),and the percentages of G2 phase cells did not change significantly. Conclusion Two different TBA could all induce Jurkat cell apoptosis and G1 phase arrest. Their inhibitory effects on tumor growth could be due to apoptosis caused by G1 arrest.
