Effects of selenium and vitamin E on myocardial fibrosis in rats fed on grains from Keshan disease endemic area

Abstract:

Objective To observe the  effects of adequate selenium (Se) and vitamin E (VE) on  myocardial fibrosis and remodelling and  explore theeffects of Se and VE in the repair process after myocardial injury in Keshan disease  area. Methods Thirty Wistar rats were randomly divided into control group (fed on mixed grain),Keshan disease area diet group (EG,fed on Keshan disease area diet continuously),Keshan disease area diet plus Se group (EG+Se),Keshan disease area diet plus VE group (EG+VE) and Keshan disease area diet plus Se and VE group (EG+Se+VE). All rats were sacrificed after 21 d. The activitiy of glutathione peroxidase(GSH-Px) was detected by colorimetry;the expression of malondialdehyde(MDA) in the serum of rats was detected by TBA. The deposition of extracelluar matrix in different groups was detected by mass on trichrome staining. The expressions of TGF-β and CTGF in myocardial cells of rats were detectedby immunohistochemical staining and Western blotting method. Results In the myocardial fibsis rats fed on Kashan disease area diet,the serum GSH-Px activity was decreased,the level of serum MDA was increased,myocardial fabric accumulation and  the expression levels of TGF-β and CTGF were up-regulated.Compared with EG group, the serum GSH-Px activities wre increased (P<0.01 and P<0.05), the MDA contents were decreased(P<0.05),the cardiac extracellular matrix deposition and the expressions of TGF-β and CTGF in rat myocardium were decreased  (P<0.01) in EG+Se group and EG+Se+VE group. Conclusion Low Se and VE play a continuing role in the repair process after myocardial injury. The adequate supplementation of Se and VE isable to reverse the myocardial fibrosis and remodelling.
