Primary culture and identification of miniature pig periodontal ligament cells

Abstract:

Objective To isolate and culture miniature pig periodontal ligament cells in vitro, pave the way for developing a animal model of miniature pig and provide  experimental basis for periodontal regeneration.Methods 4 incisors and 4 canines were extracted in the condition of asepsis.The primary cells were isolated from miniature pigperiodontal ligament and cultivated by tissue explant culture technique,then passaged when the cells completely covered the bottom of culture flasks. The growth curve was drawn. Morphological analysis and immunocytochemical analysis were used to characterize the cell lineage. Results The miniature pig periodontal ligament cells of primary culture were obtained with tissue cultivation,the success rate of primary culture was 50% and the highest successful rate (100%) was observed in incisor.The actively proliferating cells showed long spindle appearance.By immunocytochemical analysis,these cells were positive to antibodies against vimentin,and negative to antibodies against ceratin,indicating themselves to be mesoderm-derived fibroblasts .Conclusion The miniature pig periodontal ligament cells,which can be cultived successfully in vitro,especiallythe cells from incisor,may be the potential cells for periodontl regeneration in animal model.
