Abstract:Objective To observe the effects of the magnetic c-erbB2 antisense oligodeoxynucleotide probe on morphology and expression of SK-Br-3 cancer cells in vitro,and to discuss whether the probe has the functions of diagnosis and treatment. Methods SK-Br-3 cancer cells were transfected by magnetic c-erbB2 antisense probe as experiment group,Fuda tumor cells were transfected by magnetic c-erbB2 antisense probe,SK-Br-3 cancer cells were transfected by SPIO and ASODN,untransfected SK-Br-3 cancer cells as 4 control groups. The iron distribution in the transfected cancer cells was observed by Prussian blue staining,the ultrastructure was observed by transmission electron microscope,the protein expression of cerbB2 and the iron content were detected and MR imaging was performed. Results The iron particles scattered in the cytoplasm of transfected SK-Br-3 cancer cells under light microscope.The iron particles,groups-like pinocytosis, cell membrane vacuolization,cell shrinkage,nuclear condensation,and apoptotic bodies were found under transmission electron microscope,but the cells in control groups had no the changes mentioned above.Compared with control groups,the inhibitory rate of protein expression of SK-Br-3 cells was significantly increased,the iron content of cells was increased(P<0.05),the signal in MR imaging was decreased. Conclusion Magnetic c-erbB2 antisense probe can smoothly go into the SK-Br-3 cancer cells,it not only results in SK-Br-3 cell apoptosis but also can be found by MR imaging,it may become a bifunctional probe with significance in treatment and diagnosis.
