Cloning and expression of megakaryocyte protein tyrosine phosphatase 2  and its relationship with osteoporosis


Abstract: To research the cloning and expression of the megakaryocyte protein tyrosine phosphatase 2(PTPMEG2) and its regulatory role in osteoporosis development.Methods
The purified PTPMEG2 was used to prepare PTPMEG2 polyclonal antibody.30 female SD rats were rardomly divded into model group(n=15) and control group(n=15).The retinoic acid gavage method was used to construct rat osteoporosis models.The rats in control group were gavaged with saline.The acid phosphatase (ACP) and alkaline phosphatase (ALP) levels in rat serum in two groups were detected.The expression levels of PTPMEG2 in some rat tissues were detected by Western blotting method.The expression level of PTPMEG2 in bone marrow cells (BMCs),red blood cells and bone marrow stromal cells (BMSCs)were detected and compared.Results The PTPMEG2 polyclonal antibody was successfully prepared.The levels of serum ACP and ALP of rats in model group were higher than those in control group (P<0.01).The expression level of PTPMEG2 in kidney tissue of rats in model group was higher than that in control group(P<0.05),but the expression level in muscle tissue of rats in model group was lower than that in control group(P<0.05).Compared with control group,the expression levels of PTPMEG2 in red blood cells and BMCs of rats in model group has no significant changes,but the expression level of PTPMEG2 in rat BMSCs in model group was significantly increased. Conclusion The expression level of PTPMEG2 in BMCs of osteoporosis rats is increased,and mostly in mature bone cells differentiated in bone marrow;and  PTPMEG2 has important role in osteoporosis development.
