Anti-tumor effect of extract from |Bark of Juglans |Mandshurica Maxim  on gastric cancer mice in vivo

Abstract:

Abstract:Objective To study the anti-tumor activity and  effect on immune function of extract from  Bark of Juglans  Mandshurica Maxim (HYT)in orthotopic transplantation of gastric cancer mice and illustrate its underlying mechanism and  provide theoretical basis for  developing traditional low-toxicity and high-performance anti-tumor Chinese medicine and herbs in clinic.  Methods The orthotopic transplantation of gastric cancer mouse models were established with HYT.The mice were divided into high dose of HYT group,lower dose of HYT group,sham operation group,negative control group and positive control group(5-FU). The inhibitory rate,thymus index and spleen index were observed. ELISA method and flow cytometry were used to detect the IL-4,IL-12,TNF-α levels and T-lymphocyte subsets. Results The difference of the inhibitory rates between high dose of HYT group and positive control group(5-FU)has no sinificant difference(P>0.05);compared with negative control group,the thymus index and spleen index were increased(P<0.05),the IL-12 level was increased(P<0.05),the IL-4 level was decreased(P<0.05),theTNF-α level was also increased(P<0.05),the   CD4+/ CD8+ ratio of  T lymphocyte subgroup was increased(P<0.05) in high dose of HYT group. Conclusion HYT can enhance immune function and inhibit tumor growth in vivo, and it can be used as  low toxicity and high-performance anti-tumor Chinese medicine and herbs.
