Extraction of protein components from Jilin Shuangyang sika deer pilose antler and their effects on pathomorphology of major organs in mice

Abstract:

Abstract:Objective To evaluate the  security of application of the protein components from Jilin Shuangyang sika deer antler pilose and provide experimental basis for study on its development and application.  Methods
The raw pilose antler from Shuangyang was homogenated with high speed. The supernatant was obtained by centrifuge.  Ammonium sulfate was applied to precipitate protein solution and then PBS was used to dissolve. Sephadexg-25 was applied to eliminate irons to obtain purified total protein solution. 58 ICR mice  were randomly divided into control group(saline intragastric administration),intragastric group,subcutaneous injection group,and intraperitoneal injection group all by administrating protein solution. The dosage was 6.7 mg?kg-1,twice a day. 28 days later,the mice were executed and the heart,liver,kidney,lung of  each group were obtained for  pathological observation. Results After sephadex-G25 filtration,the concentration of total protein solution was 0.685 g?L-1 . Myoplasm condensation was found in  intragastric group(50.0%),subcutaneous injection group(91.7%) and intraperitoneal injection group(87.5%). Inflammatory cell infiltration and focalnecrosis were observed in intragastric injection group(40.0%),subcutaneous injection group (64.3%) and intraperitoneal injection group(77.8%). 3 cases of hyaline degeneration were found in subcutaneous injection group. Every 3 cases of pneumonia were found in intragastric injection group.Every 3 cases of broaden pulmonary septum were found in subcutaneous injection group and intraperitoneal injection group. Conclusion Antler protein of sika deer can result in toxication of heart and liver of mice in some extent, but has no  significant effect on either kidney or lung.
