Influence of blocking FAK expression in invasion of salivary adenoid cystic carcinoma cells

Abstract:

Abstract:Objective To study the influence of blocking the expression of focal adhesion kinase(FAK) in the invasion behaviorof salivary adenoid cystic carcinoma-lung metastasis(SACC-LM)cells and explore its relevant mechanism.Methods The SACC-LM cells in logarithmic growth phase were selected,and randomly divided into blank control group,DMSO groupand Genistein group,the Transwell chamber was used to observe the influence of Genistein in the invasion of SACC-LM cells.TheSACC-LM cells were divided into blank control group,different doses of  Genistein groups and Genistein groups for different treatment time,the expressions of FAK and extracellular signal regulated kinase(ERK) were detected withWestern blotting.Results Compared with bland control group,the invasive ability of SACC-LM cells was reduced  and the expressions of FAK andERK protein were decreased with the increase of the inhibitor concentration and prolongation of treatment time (P<0.05) inGenistein groups.Conclusion Blocking   FAK expression can reduce the invasion of SACC-LM cells,the changes of FAK affect theexpression of ERK protein in SACC-LM cells.
