子宫颈鳞癌组织中PARP-1、Caspase-3、survivin和Bax的表达及其临床意义

Expressions of PARP-1,Caspase-3,survivin and Bax in cervical squamous cell carcinoma tissues and their clinical significances

摘要: 目的:探讨聚ADP核糖聚合酶-1(PARP-1)、半胱氨酸天冬氨酸蛋白酶3(Caspase-3) 、存活素(survivin)和Bcl-2家族的前凋亡蛋白(Bax)在子宫颈鳞癌组织中的表达及其与宫颈癌临床病理特征的关系,为宫颈癌早期诊断和治疗奠定实验基础。方法:采用免疫组织化学技术检测60例子宫颈鳞癌、40例宫颈原位癌和20例癌旁正常宫颈上皮(NCE)标本中PARP-1、Caspase-3、survivin和Bax的表达率,分析其表达与宫颈鳞癌的临床分期、病理类型及有无淋巴结转移的关系。结果:PARP-1在子宫颈鳞癌、原位癌和NCE中的阳性表达率分别为68.33%、62.50%和25.00%;Caspase-3为43.33%、47.50%和70.00%;survivin为95.00%、92.50%和5.00%;Bax为31.67%、32.50%和75.00%。PARP-1和survivin在子宫颈鳞癌组及宫颈原位癌组阳性表达率低于NCE组(P<0.01);Caspase-3和Bax在子宫颈鳞癌组及宫颈原位癌组阳性表达率高于NCE组(P<0.01)。PARP-1、survivin和Bax的阳性表达率与临床分期、病理类型和有无淋巴结转移均有关联(P<0.01);而Caspase-3阳性表达率与上述指标无关联(P>0.01)。子宫颈鳞癌组织中PARP-1与Caspase-3、survivin与Bax表达阳性率均呈负相关关系(r=-0.71,P=0.043;r=-0.63,P=0.038)。结论:PARP-1、survivin在子宫颈癌组织中表达水平上调和Caspase-3、Bax表达水平下调可能在子宫颈鳞癌浸润和转移中起重要作用。

Abstract: Objective To explore the expressions of poly ADP-ribose polymerase(PARP-1),cysteinyl aspartate specific proteinase(Caspase-3),survivin and Bax in cervical squamous cell carcinoma tissues and their relationships with the  clinicopathogical characteristics of cervical carcinoma,and to provide experimental basis for early diagnosis and  treatment for cervical carcinoma.Methods Immunohistochemical method was used to detect the expression rates of PARP-1,Caspase-3,survivin and Bax in specimens of 60 cases of cervical squamous cell carcinoma,40 cases of cervical carcinoma in situ and 20 cases of  normal cervical epithelium (NCE) adjacent to carcinoma.The relationships between their expression levels and clinical stages,pathological types and lymph node metastasis or not were analyzed.Results The positive expression rates of PARP-1 in cervical squamous cell carcinoma,cervical carcinoma in situ and NCE tissue were 68.33%,62.50%,and 25.00%,respectively;the positive expression rates of Caspase-3 in cervical squamous cell carcinoma,cervical carcinoma in situ and NCE tissue were 43.33%,47.50%,and 70.00%, respectively;the positive expression rates of survivin  were 95.00%,92.50%,and 5.00%,respectively;the positive expression rates of Bax  were 31.67%,32.50%,and 75.00%,respectively.The positive expression rates of PARP-1 and survivin    in  squamous cell carcinoma and carcinoma in situ groups were lower than those in NCE group (P<0.001);the positive expression rates of Caspase-3 and Bax  in  squamous cell carcinoma and carcinoma in situ groups were higher than those in NCE group (P<0.01).The  positive expression rates  of  PARP-1,survivin and Bax had relationships  with the clinical stage,histological type,and lymph node metastasis(P<0.01);but the positive expression rate of  Caspase-3 didn’t have relationship with those indexes mentioned above(P>0.01).The  positive expression rates of PARP-1 and Caspase-3 in cervical squamous cell carcinoma tissue were negatively correlated (r=-0.71,P=0.043);the positive  expression rates of  survivin and Bax in cervical squamous cell carcinoma tissue  were negatively correlated(r=-0.63,P=0.038).Conclusion The up-regulation  of the expression levels  of PARP-1 and survivin and the down-regulation of the levels of Caspase-3 and Bax expression are likely to play an important role in the invasion and metastasis of cervical squamous cell carcinoma.
