摘要: 目的:研究三氯化镧(LaCl3)对大鼠肝癌细胞生长的影响。 方法:以体外培养大鼠肝癌细胞株CBRH-7919为研究对象,给予LaCl3(0.01、0.10及1.00 mmol•L-1)培养不同时间(1、3和5 d)后,观察CBRH-7919细胞生长、集落形成及细胞学变化。同时,运用流式细胞术检测CBRH-7919细胞周期变化。 结果:0.10 mmol•L-1 LaCl3组培养3 d,对CBRH-7919生长具有明显的抑制作用(P<0.01),随着LaCl3剂量增加和培养时间延长,对该细胞生长的抑制作用明显增强(P<0.01);0.1、1.00 mmol•L-1 LaCl3组的集落形成抑制率同对照组相比,差异均有显著性(P<0.01)。CBRH-7919细胞经LaCl3作用后,G0/G1期细胞百分数增加,0.01、0.10和1.00 mmol•L-1 LaCl3组与对照组相比,差异均有显著性(P<0.01);S期细胞百分数减少,0.10、1.00 mmol•L-1 LaCl3组与对照组相比,差异均有显著性(P<0.05)。 结论:LaCl3对CBRH-7919细胞生长具有明显的抑制作用。

Abstract: Objective To study the effect of LaCl3 on the growth of CBRH-7919 cells. Methods The growth, clone inhibitory rate and general histological changes of CBRH-7919 cells were observed at different time (1th,3rd and 5th day) after treated with different doses (0.01,0.10 and 1.00 mmol•L-1) of LaCl3 in vitro. At the same time, the changes of cell cycle were detected by flow cytometry. Results The evident inhibition of LaCl3 to CBRH-7919 cells growth occurred at 3rd day after treatment (P<0.01), gradually strengthened following the increase of dose and time (P<0.01). The result of clone formation assay expressed that the clone formations of CBRH-7919 cells were inhibited distinctly at the concentration of 0.10 and 1.00 mmol•L-1 LaCl3 (P<0.01). The result indicated that treated by LaCl3, the distribution of cell cycle changed as followed: the cell percentage of G0/G1 period was increased, the ratios of groups of 0.01, 0.1 and 1.00 mmol•L-1 LaCl3 were evidently increased compared with the control and presented density-dependent relation (P<0.01); The cell percentage of S period was reduced, the rations of groups of 0.10 and 1.00 mmol•L-1 LaCl3 were significantly reduced compared with the matched control (P<0.05). Conclusion LaCl3 can inhibit the growth of CBRH-7919 cells obviously.
