TIMP-3对前列腺癌PC-3M细胞株增殖和侵袭能力的影响

Effects of TIMP-3 on proliferation and invasion of human prostate carcinoma PC-3M cell line

摘要: 目的:观察TIMP-3对前列腺癌细胞PC-3M增殖和侵袭能力的影响。 方法:实验分为前列腺癌细胞PC-3M组、转染空质粒PCDNA3.1的PCDNA-PC-3M组和转染 TIMP-3的PCDNA-TIMP-3-PC-3M组。采用黏附实验、MTT比色法对比观察稳定转染的PCDN A-TIMP-3-PC-3M与PCDNA-PC-3M的黏附能力和增殖活力。采用单层细胞侵袭和Boyden 小室侵袭实验,对比观察TIMP-3对PC-3M细胞体外侵袭能力的影响。 结果:稳定转染TIMP-3的PC-3M细胞的黏附率和细胞增殖速度明显低于空质粒组和未 转染 组(P<0.05),稳定转染TIMP-3的PC-3M细胞的侵袭指数明显低于空质粒组和未转染 组(P<0.05),空质粒组和未转染组间细胞增殖速度和细胞侵袭指数比较差异无显著 性(P>0.05)。 结论:TIMP-3对前列腺癌PC-3M细胞增殖和侵袭能力具有抑制作用。

Abstract: Objective To assess the effects of tissue inhibitor of metalloproteinase-3 (TIMP-3) on proliferation and invasion of PC-3M cell line in vitro. Methods The adhesion assay was used to detect the effects of TIMP-3 on cancer cell adhesion abilities. MTT colorimetry was used to detect the proliferation abilities of PC-3M cells. Monolayer and Boyden chamber invasion assay were performed to assess cell invasion abilities. Results The adhesion rate and proliferation rate of transfected cells containing TIMP-3 gene were significantly lower than those of cells transfected with empty plasmid and untransfected cells (P<0.05). The cell invasion number and index of the former were significantly lower than the others (P<0.05). The differences of adhesion rate, proliferation rate and invasion index between transfected empty plasmid cells and untransfected cells weren′t statistically significant (P>0.05). Conclusion TIMP-3 can significantly inhibit the proliferation and invasion of human prostate carcinoma PC-3M cell line.
