外源硫酸锌对糖尿病大鼠肝脏损伤的保护作用

Protective effect of exogenous zinc sulfate on liver injury of diabetic rats

摘要:

目的:观察外源性硫酸锌对链脲佐菌素(STZ)诱导糖尿病大鼠模型肝脏组织损伤的保护作用并探讨其机制。方法:健康Wistar大鼠50只,随机分为正常对照组(n=9)、高脂对照组(n=10)、高脂糖尿病模型组(n=7)、高脂糖尿病锌干预组(n=8)和高脂糖尿病后锌干预组(n=8)。锌干预组以锌离子浓度15 mg?kg-1?d-1进行灌胃,其他组以相应体积蒸馏水灌胃。糖尿病模型建立8周末,测定肝功,计算肝脏指数,测定肝脏组织超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-Px)。结果:与糖尿病模型组比较,外源性硫酸锌干预能明显降低糖尿病大鼠的肝脏指数(P<0.05);血清AST、ALT 水平有下降趋势(P>0.05);减少肝脏组织MDA水平,并显著增加SOD及GSH-Px水平(P<0.05);锌干预组组间肝脏组织MDA及SOD水平比较差异有显著性(P<0.05)。结论:外源性硫酸锌能够降低糖尿病模型大鼠肝脏指数,并改善其肝功能损伤,其机制可能与硫酸锌改善肝脏过氧化状态有关。

Abstract:

Abstract:Objective
To study the protective effect of zinc sulfate on liver injury of diabetic rats induced by STZ and explore its mechanism. Methods Fifty  male Wistar rats were randomly divided into five groups: normal diet (NC) group, high fat diet (FC) group, diabetes (DM) group, Zn before the intervention of diabetes (ZnDM) group, Zn after the intervention of diabetes (DMZn) group. The rats in Zn groups were fed with Zn ions (Zn2+) 15 mg?kg-1?d-1 by giving ZnSO4 solution daily by gavage, the others were fed with distilled water by gavage. Eight weeks after STZ injection, the serum was collected to detect the function of liver, AST and ALT, and liver was taken for the measurement of liver index, SOD, MDA, and GSH-Px. Results  Compared with DM group, Zn intervention could significantly reduce the liver index (P<0.05); the levels of AST and ALT in serum were decreased mildl (P>0.05); the level of MDA in liver tissues was markedly decreased and the levels of SOD and GSH-Px in liver tissue were significantly improved (P<0.05); the levels of MDA and SOD in liver tissues had significant differences between the two groups of Zn intervention (P<0.05). Conclusion Exogenous zinc sulfate can decrease the liver index of diabetic rats, and make the liver function better. The mechanism of the protection of Zn may be related to the strengthening of anti-oxidation.
