胞二磷胆碱对1-甲基-4-苯基吡啶诱导的多巴胺能神经元损伤的保护作用

Neuroprotective effect of citicoline on dopaminergic neuron injury induced  by MPP+ in mouse mesencephalic dissociated culture

摘要: 目的:探讨胞二磷胆碱对1-甲基-4-苯基吡啶(MPP+)诱导的多巴胺能神经元数量减少及形态学改变的影响。方法:体外原代培养怀孕第14 天小鼠胚胎中脑神经元,采用MPP+(10 μmol•L-1)在体外建立多巴胺能神经元损伤细胞模型。实验分为对照组、单纯MPP+药物组及5个不同浓度胞二磷胆碱(0.01、0.101.00、10.00和100.00 μmol•L-1)与MPP+共同给药组。应用酪氨酸脱氢酶免疫化学染色方法观察不同浓度胞二磷胆碱对MPP+诱导多巴胺能神经元损伤的形态及数量的变化。结果:体外培养第12天,单纯MPP+药物组中多巴胺能神经元的数量明显少于对照组(P<0.05);0.10μmol•L-1和100.00μmol•L-1 胞二磷胆碱治疗组,多巴胺能神经元的数量分别高于单纯MPP+药物组(11.83%和21.26%)(P<0.05)。0.10和100.00 μmol•L-1胞二磷胆碱加MPP+药物组中多巴胺能神经元的突起长度明显高于单纯MPP+药物组(P<0.05)。结论:胞二磷胆碱可有效地防止MPP+的多巴胺能神经元的损伤和死亡,可能为帕金森氏病的防治提供一个新途径。

Abstract: To investigate the effect of citicoline on dopaminergic neuron injury induced by MPP+. Methods E14 mouse embryos were used to make mecencephalic dissociated culture.10 μmol•L-1 MPP+ was used to set up the cell model of damage of dopaminergic neurons.The cell cultures were divided into control group,MPP+- trerated group,and another five citicoline (with different concentrations of 0.01、0.10、1.00、10.00 and 100.00 μmol•L-1,respectively) and MPP+ -cotreated groups. Tyrosine hydrolysis (TH) immunochemical stainning was used to identify dopaminergic neurons.The morphological changes and the number of dopaminergic neurons were evaluated. Results On the 12th day in vitro(DIV),the number of TH-ir (TH immunal reactive) neurons was significantly reduced by MPP+ (P<0.05).The number of TH-ir neurons increased by 11.83% and 21.26% of control level when treated with 0.10 or 100.00 μmol•L-1 of citicoline for 6 d respectively,compared with MPP+-treated alone group (P<0.05).The average length of the longest denrite of dopaminergic neurons was much longer in 0.01 μmol•L-1and 100.00 μmol•L-1 citicoline cotreated group,compared with MPP+-treated alone group(P<0.05).Conclusion Citicoline could effectively prevent the death and morphological changes of dopaminergic neurons induced by MPP+,and provide a new treatment way for Parkinson’s dis ease (PD.
