抗人成骨肉瘤单克隆抗体的制备及其特性

摘要: 目的:制备抗人成骨肉瘤单克隆抗体并探讨其特性。方法:用人骨肉瘤细胞系OS-732免疫BALB/c小鼠,取其脾细胞与骨髓瘤细胞系NS-1融合,经酶联免疫吸附试验及补体依赖细胞毒实验筛选,获3株分泌抗人成骨肉瘤单克隆抗体(McAb)的杂交瘤细胞。将手术切取经病理证实的骨肉瘤、软骨肉瘤、骨巨细胞瘤、滑膜肉瘤、平滑肌肉瘤、非骨化性纤维瘤、骨软骨瘤和正常肌、骨组织冰冻切片后,应用间接免疫荧光抗体实验检测。 结果:3株McAb均与骨肉瘤组织呈阳性反应,其中二株亦与软骨肉瘤呈阳性反应,与正常组织及其他肿瘤组织呈阴性反应。3株McAb分别与骨肉瘤相关抗原不同的抗原决定簇结合。 结论:抗人成骨肉瘤单克隆抗体具有较高的特异性。

Abstract: Objective To prepare anti-osteosarcoma monoclonal antibodies (McAb) and study its characteristics. Methods Splenocytes of BALB/c mice immunized with osteosarcoma cell line OS-732 were extracted and these cells were fused with murine myeloma cell line NS-1. Enzyme-linked immunosorbent assay (ELISA) and complement-dependent cytotoxic assay were used to screen and three hybriddoma cell lines capable of screting antiosteosarcoma monoclonal antibodies were acquired. The tissues of osteosarcoma, chondrosarcoma, giant cell tumor of bone,leiomyosarcoma, synoviosarcoma, nonossifying fibroma,osteochondroma, normal tissues of muscle and bone were made into frozen slides and the indirect immunofluorescence test were applied to test these specimens. Results All the three lines showed positive results in the osteosarcoma slides and two of them showed weakly positive results in the chondrosarcoma. The three McAb lines combined with different antigenic determinant epitope of osteosarcomous correlative antigens, respectively. Conclusion McAb possesses higher sensitivity and specificity.
