低剂量博来霉素A5促进荷瘤小鼠化疗效果及其机制探讨

Antitumor effect of low dose BLMA5 and its mechanisms

摘要: 目的:研究低剂量博来霉素 A5(BLMA5,0.08 mg•kg-1)诱导的适应性反应对皮下接种S180肉瘤的昆明种小鼠模型常规化疗效果的影响及其机制。方法:荷瘤小鼠化疗前预先给予低剂量BLMA5(D1)处理,观察肿瘤生长的动态, 制备小鼠骨髓细胞染色体标本及测定肝组织自由基活性。 结果:荷瘤小鼠化疗前预先给予D1处理可使肿瘤生长明显减慢(P<0.05~0.001),骨髓细胞染色体畸变减轻(P<0.01),肝脏SOD活性提高(P<0.05),GSH-Px活性也有增高趋势,且MDA含量降低(P< 0.01)。结论: 低剂量BLMA5提高化疗效果的机制可能与改善机体抗氧化能力及提高自由基产物的清除能力有关。

Abstract: Objective To study the common chemotherapy effect and its mechanisms of the adaptive response induced by low dose BLMA5 (0.08 mg•kg-1, D1) on a Kunming S180 sarcoma mouse model made through subcutaneous injection. Methods D1 was given to tumor-bearing mice before chemotherapy, the growth of tumor was observed, chromosomal samples of bone marrow cells of mice were prepared and the activities of free radicals of liver were measured. Results The grow velocity was significantly decreased (P<0.05~0.001), the chromosome aberrations of bone marrow cells were significantly reduced (P<0.01), SOD activity of liver was increased markedly (P<0.05), the activity of GSH -Px also had the trend of increasing and the content of MDA decreased significantly (P<0.01) when D1 was given to the tumor-bearing mouse before larger dose BLMA5(D2) chemotherapy. Conclusion The chemotherapy effect enhanced with low dose BLMA5 is associated with the increase of antioxidant capacity and the elimination capacity of free radicals products.
