黄芪注射液对糖尿病大鼠肾脏的保护作用及其机制

Protective effect of Injection of Milkvetch Root on kidney of diabetic rats and its mechanism

摘要:

目的:观察黄芪注射液对大鼠血脂代谢、自由基氧化损伤和肾功能等方面的影响,探讨其对2型糖尿病大鼠肾脏的保护机制。方法:将80只 Wistar 大鼠随机分为对照组、模型组、卡托普利组和黄芪注射液组,每组20只。 除对照组外,其他各组大鼠每天给予高糖高脂饮食,8周后腹腔注射小剂量链脲佐菌素(STZ)1次建立2型糖尿病模型。黄芪注射液组大鼠于造模成功后第17周开始每天腹腔注射黄芪注射液 2 mL•kg-1,4 周后检测各组大鼠空腹血糖(FBG)、血清总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-c)、低密度脂蛋白胆固醇(LDL-c)、尿微量白蛋白(Ualb)、肌酐(Scr)、尿素氮(BUN)水平、尿蛋白排泄率(UAER)和肌酐清除率(CCr),检测肾脏组织中丙二醛(MDA)水平、超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-Px)活性。结果:与对照组比较,模型组大鼠FBG、TC、TG、LDL-c、MDA、UAlb、BUN、SCr和UAER升高(P<0.05或P<0.01),HDL-c水平、SOD和GSH-Px活性、CCr降低(P<0.05或P<0.01)。与模型组比较,黄芪注射液组大鼠血清TC、TG和LDL-c水平,肾组织MDA水平,UAER和血清UAlb、BUN、SCr水平均显著降低(P<0.05或P<0.01),SOD和GSH-Px 活性及CCr显著升高(P<0.05或P<0.01),HDL-c水平无明显变化。结论:黄芪注射液能纠正2型糖尿病大鼠的脂质代谢紊乱,抵抗自由基介导的脂质过氧化,对糖尿病大鼠的肾脏有保护作用。

Abstract:

To observe the effects of Injection of Milkvetch Root on blood lipid metabolism,free radical oxidative damage and renal function of rats,and to discuss the  protective mechanism of injection of Milkvetch Root  on kidney of  type 2 diabetic rats.Methods 80 Wistar rats were randomly divided into control group,model group,captopril group,and injection of Milkvetch Root(IMR) group;there were 20 rats in each group.Except for control group,the rats in the other groups were given high sugar and high fat diet for 8 weeks,and the rats were intraperitoneally injected with low dose of streptozocin(STZ) once to induce type 2 diabetic rat models.The rats in IMR  group were  intraperitoneally injected with Injection of Milkvetch Root (2 mL•kg-1) since the 17th week after establishment of models.The levels of fasting blood glucose(FBG),serum total cholesterol (TC),triglyceride (TG),high-density lipoprotein cholesterol (HDL- c),low density lipoprotein cholesterol (LDL-c),urine trace albumin (Ualb),creatinine(Scr),content of urea nitrogen (BUN), urinary albumin excretion rate (UAER),and creatinine clearance rate (CCr),the content of malondialdehyde (MDA) in kidney tissue,and the activities of superoxide dismutase (SOD) and glutathione peroxidase (GSH-px) of the rats in various groups were detected after 4 weeks.Results  Compared with control group,the levels of FBG,TC,TG,LDL-c,MDA,UAlb,BUN,SCr and UAER were increased;the  HDL-c level,CCr and the activities of SOD and GSH-Px were decreased(P<0.05 or P<0.01).Compared with model group,the levels of serum  TC,TG and LDL-c and level of MDA in kidney tissue,UAER and serum UALb,BUN,and Scr of the  rats in IMR group were significantly decreased(P<0.05 or P<0.01);the activities of serum SOD  and GSH-Px and  CCr  were increased markedly (P<0.05 or P<0.01);the HDL-c level had no significant change.Conclusion   Injection of Milkvetch Root can correct the lipid metabolic disorders and resist free radical mediated lipid peroxidation in the  type 2 diabetic rats to protect their kidneys.
