北五味子提取物对实验性肝纤维化大鼠肝损伤的保护作用

Protective effects of extract fructus Schisandrae Chinensis on hepatic injury in experimental hepatic fibrosis rats

摘要:

目的:观察北五味子提取物(EFSC)对实验性肝纤维化大鼠肝损伤的保护作用,并探讨其作用机制。方法:将60只Wistar大鼠随机分为对照组,模型组,马洛替脂组,100、200和400 mg/kgEFSC组,每组10只。皮下注射CCl4花生油溶液10周建立大鼠实验性肝纤维化模型。各组大鼠给药4周后测定血清丙氨酸转氨酶(ALT)和天冬氨酸转氨酶(AST)活性,白蛋白(ALB)、球蛋白(GLO)、总蛋白(TP)和一氧化氮(NO)水平,检测各组大鼠肝组织中超氧化物歧化酶(SOD)和一氧化氮合酶(NOS)活性,丙二醛(MDA)、谷胱甘肽(GSH)和羟脯氨酸(Hyp)水平。结果:与模型组比较,200和400 mg/kgEFSC组大鼠血清ALB水平和ALB/GLO比值显著升高(P<0.05或P<0.01),血清AST、ALT活性和GLO、NO水平均显著降低(P<0.05或P<0.01);肝组织中SOD活性和GSH 水平显著升高(P<0.05或P<0.01),Hyp和MDA水平显著降低(P<0.05)。结论:EFSC可改善肝功能,抑制肝脏胶原纤维的生成,纠正肝纤维化引起的ALB水平低下,减轻肝细胞脂质过氧化损伤,从而防止肝纤维化的形成与发展。

Abstract:

Abstract:Objective
To study the protective effects of extract fructus Schisandrae Chinensis (EFSC) on hepatic injury in experimental hepatic fibrosis rats,and to clarify its mechanism.Methods 60  Wistar rats were randomly divided into control group(n=10),model group(n=10),malotilate group(n=10),and 100,200 and 400 mg/kg EFSC groups(n=10).The experimental hepatic fibrosis model was established by hypodermic injection with CCl4  arachis oil solution for 10 weeks.The activities of aspartate aminotransferase (AST) and alanine aminotransferase (ALT),and the levels of albumin (ALB),globulin (GLO),total protein (TP) and nitrogen monoxidum (NO) in serum of the rats in various groups were determined;the activities of superoxide dismutase (SOD) and nitricoxide synthase (NOS),and the levels of malonaldehyde (MDA),glutathion (GSH) and hydroxyproline (Hyp) in liver tissue of the rats in various groups were detected.Results Compared with model group,after treated with  200 and 400 mg/kg  EFSC  for 4 weeks,the levels of ALB and ALB/GLO ratio in serum of the rats were significantly increased (P<0.05 or P<0.01);the activities of AST and ALT and the  levels of GLO and NO in serum of the rats were significantly decreased (P<0.05 or P<0.01);the activities of SOD and the levels of GSH in liver  tissue of the rats were   significantly increased(P<0.05 or P<0.01) and the levels of MDA and Hyp in liver tissue of the rats were significantly decreased(P<0.05).Conclusion EFSC can improve the hepatic function,inhibit the generation of hepatic collagen fiber,correct the low level of ALB and lighten the damage of lipid peroxidation in hepatocytes,so as to prevent formation and development of hepatic fibrosis.
