吗啡对肠道嘌呤核苷酸分解代谢的影响

Effect of morphine on purine nucleotides catabolism in intestine

摘要: 目的:研究吗啡依赖及戒断状态下嘌呤核苷酸分解代谢的变化及作用机制。方法:剂量递增腹腔注射盐酸吗啡,建立吗啡依赖及戒断大鼠模型。试剂盒检测血浆尿酸含量、血浆及小肠组织黄嘌呤氧化酶(XO)的活性。提取小肠组织总RNA,采用逆转录聚合酶链反应(RT-PCR)检测小肠组织XO mRNA的水平,以β-actin mRNA为内标。结果:血浆尿酸水平显示,7 d吗啡用药组明显高于对照组(P<0.05);血浆XO酶学结果显示,7 d吗啡用药组XO明显高于对照组(P<0.05),自然戒断组与纳洛酮对照组也明显高于对照组(P<0.01);小肠组织XO酶学结果可见, 7 d吗啡用药组与纳洛酮戒断组XO明显高于对照组(P<0.05);小肠组织XO的mRNA水平可见,7 d吗啡用药组、自然戒断组和纳洛酮戒断组XO mRNA水平明显高于对照组。结论 :吗啡促进嘌呤核苷酸分解代谢;吗啡促进大鼠小肠嘌呤核苷酸分解代谢关键酶XO的活性,纳洛酮不能阻断该作用。提示吗啡对小肠XO的作用是通过非μ受体途径。

Abstract: Objective To investigate the changes of purine nucleotides catabolism on morphine-dependent and withdrawal condition and its mechanism. Methods Rats were administered with morphine by intraperitoneal injection with increasing doses to establish addiction models.The level of uric acid in plasma and the activities of xanthine oxidase (XO) in the plasma and the small intestine were determined. RT-PCR was used to examine the gene transcripts of key enzyme of purine nucleotides catabolism ,XO mRNA. β-actin was used as control gene in RT-PCR study.Results The level of uric acid in plasma of morphine-administered group was significantly higher than that of control group (P<0.05).The activity of XO in plasma of morphine-administered group was higher than that of control group (P<0.05),as well as natural withdrawal group and naloxone control group (P<0.01). The activities of XO in the small intestine of morphine-administered group and naloxone withdrawal group were extremely higher than that of control group (P<0.05). The levels of transcript of XO mRNA of morphine-administered group, natural withdrawal group and naloxone withdrawal group were higher than that of control group. Conclusion Morphine can promote the purine nucleotides catabolism.Morphine may increase the activity of the key enzyme of purine nucleotides catabolism in the small intestine, which is related to the increasing of gene expression. Morphine can induce XO synthesis in mRNA level, naloxone can′t obstruct it in the short time.
