VEGF对体外培养的人脐静脉内皮细胞Ets-1的诱导表达

VEGF-induced expressions of Ets-1 of cultured human umbilical endothelial cells in vitro

摘要: 目的:探讨血管内皮细胞生长因子(VEGF)对体外培养的人脐静脉内皮细胞(HUVECs)中转录因子Ets-1的诱导表达。 方法:原代培养获得HUVECs;应用细胞免疫化学染色技术及形态计量分析系统Image-Pro Plus(IPP),检测VEGF对培养的内皮细胞Ets-1蛋白的诱导表达作用并对相关指标进行半定量分析。结果:VEGF(10 μg•L-1)可诱导内皮细胞Ets-1蛋白的短时表达升高(P<0.01),即随时间的延长,Ets-1表达也逐渐增强,在10 min时达高峰,30 min后Ets-1表达逐渐降低到无VEGF刺激的水平。 结论:VEGF可诱导人脐静脉内皮细胞中Ets-1蛋白的短时表达增加。

Abstract: Objective To study the VEGF-induced expressions of Ets-1 of human umbilical vein endothelial cells(HUVECs) cultured in vitro. Methods The HUVECs were cultured and obtained. The immunocytochemical stain and Image-Pro Plus (IPP) analysis system were used to determine the VEGF-induced expressions of Ets-1 in the cultured endothelial cells. The relative indexes were analyzed with semi-quantitative technique. Results The VEGF (10 μg•L-1) induced the short-term expressions of Ets-1 in HUVECs (P<0.01), and Ets-1 expressions increased with the prolongation of time. The expressions of Ets-1 protein in HUVECs reached a peak at 10 min. After 30 min, the intensity of the expressions of Ets-1 decreased gradually to the level of no VEGF-induced. Conclusion The VEGF can induce the short-term expressions increase of Ets-1 of HUVECs.
