缝裂层孔菌对小鼠的抑瘤及免疫调节作用

Anti-tumor and immunodulational effect of Phellinus linteus

摘要: 目的:研究缝裂层孔菌(PL)的抗肿瘤作用,初步探讨其抑瘤作用的机理。方法:建立小鼠实体瘤动物模型,连续10 d灌胃给予PL,处死后同时检测治疗组及对照组皮下瘤重、胸腺重、脾重;采用MTT法检测荷瘤小鼠脾T淋巴细胞及B淋巴细胞对ConA及LPS刺激的转化能力;采用绵羊红细胞作为抗原,检测荷瘤鼠抗体生成细胞功能;采用L929为靶细胞 ,检测NK细胞杀伤L929细胞功能;荷瘤鼠腹腔注射诱导剂诱生巨噬细胞,体外检测巨噬细胞 吞噬中性红功能。结果:PL 2.0、1.0 g•kg-1可抑制小鼠肝癌、胃癌和S180细胞的生长,其对肝癌的抑制率分别为38.19%和37.15%,对胃癌的抑制率分别为32.46%和30.91%,对小鼠S180肉瘤的抑制率分别为36.09%和34.16%,与对照组比较差异有显著性(P<0.05);PL 2.0 g•kg-1可明显提高小鼠T、B淋巴细胞对ConA及LPS的反应性,提高淋巴细胞转化功能(P<0.05);抗体生成细胞实验结果显示,PL 2.0及1.0 g•kg-1剂量组A值高于对照组,说 明其溶血红细胞释放血红蛋白量多于对照组,其抗体生成细胞溶解绵羊红细胞能力强于对照组(P<0.05, P<0.001);PL 2.0 g•kg-1也可提高小鼠NK细胞杀伤L929细胞功能, 杀伤率为67.82%,明显强于对照组(P<0.01);PL 2.0及1.0 g•kg-1还可明显增强小鼠腹腔 巨噬细胞的吞噬功能,与对照组比较差异有显著性(P<0.05,P<0.01)。结论:PL能增强免疫细胞功能 ,提高机体免疫力,其抗瘤作用与激活体内免疫系统有关。

Abstract: Objective To study the anti-tumor activity of Phellinus linteus(PL) and itsimmunomodulational effects. Methods Solid tumor was induced by injecting HepA, MFC, S180 cells (3106cells/animal) subcutaneously to the right anterior limb in mice. The tumor weight, thymus weight and spleen weight were measured after mice were executed. The effects of PL on lymphocyte proliferation, mitogen-induced proliferatoin of ICR mouse splenocytes were measured by MTT. Meanwhile, lymphocytes were prepared as described above. SRBC (10%, 0.2 mL) was injected to abdomina l cavity to elevate the level of antibody production by B cells. Spleen cells suspension (1×107•mL-1) was used as the effect cells and L929 cells was used as the target cells. L929 cells engulfing neutral red determined the cytotoxic act ivity of NK. Simultaneously, phagocytosis of mice was detected using the method that macrophages engulfed neutral red. The effect of PL on the production of antibody was studied, the lymphocytes from mice wereprepared as described above. Results The high-dose (2.0 g•kg-1) and mid-dose (1.0 g•kg-1) of PL could significantly inhibit the growth of HepA, MFC and S180tumor, the inhibitory rates were 38.19% and 37.15%,32. 46% and 30.91%,36.0% and 34.16%, respectively (P<0.05, P<0.01). The average absorbanc e of lymphocytes in 2.0 g•kg-1 group was higher than that in the untreated control group. The numbers of antibody forming cells were increased by PL treatment at doses of 2.0 g•kg-1 and 1.0 g•kg-1 (P<0.05, P<0.001), the difference was significant compare d with control group (P<0.05). PL (2.0 g•kg-1) enhanced the cytoto xic activity of NK cells (P<0.01). Average weights of the spleens and thymus (0.1 mg per gram body weight) were not significantly greater than those in positive control groups . PL(2.0 and 1.0 g•kg-1) increased the activity of macrophage (P<0.05, P<0.01). Conclusion PL has anti-tumor activity and immunomodulational effect and probably inhibit tumor by means of activating the immunocompetence of the body.
