人胶质瘤MMP-2及TIMP-2 mRNA表达和酶活性的改变及其意义

摘要: 目的:探讨人胶质瘤基质金属蛋白酶2(明胶酶A,MMP-2)及金属蛋白酶组织抑制物2(TIMP-2)mRNA表达和活性的意义。 方法:按WHO分类标准将26例人胶质瘤标本分为4组,用RT-PCR检测MMP-2和TIMP-2 mRNA表达,用酶谱法检测其活性。 结果:在明胶酶谱分析MMP-2显示两条带,分别是定位于62 000活性形式和72 000酶原形式。在各级别胶质瘤之间MMP-2的酶原形式(72 000)无差别。随胶质瘤恶性程度的增加,MMP-2 mRNA的表达水平和酶活性均明显增加,而TIMP-2 mRNA的表达水平和酶活性均随胶质瘤恶性程度的增加而明显降低。 结论:在不同恶性级别人胶质瘤,MMP-2和TIMP-2的基因转录及酶活性均发生改变,这些改变在促进胶质瘤的侵袭生长中起重要作用。

Abstract: Objective To investigate the mRNA expressions and enzymatic activities for both matrix metalloproteinase-2(MMP-2)and tissue inhibitor of metalloproteinase-2(TIMP-2)in human gliomas and their significances. Methods The samples from 26 cases of glioma were divided into four groups according to the WHO classification, normal brain samples served as the control. The mRNA expressions for both MMP-2 and TIMP-2 were determined using reverse transcriptase-polymerase chain reaction(RT-PCR)and the enzymatic activities of MMP-2 and TIMP-2 were detected by gelatin zymography in the samples. Results In gelatin zymography, MMP-2 revealed two bands, which were localized at 62 000 (activated form) and 72 000 (latent zymogen form) of MMP-2, respectively. The latent zymogen form(72 000) of MMP-2 was not different among the four grades of gliomas. The mRNA expression level and the enzymatic activity of MMP-2 were significantly elevated with the malignant grade of the gliomas, while the mRNA expression level and the activity of TIMP-2 were markedly reduced with increasing the malignant grade of the gliomas. Conclusion The gene transcription and the enzymatic activities for both MMP-2 and TIMP-2 have changed in human gliomas with different grades of malignancy, which play an important role in the facilitation of glioma invasion.
