异氟醚预处理对缺血再灌注损伤后幼兔心肌TNF-α和iNOS mRNA表达的影响及其机制

摘要:

的:观察异氟醚(ISO)预处理对幼兔缺血再灌注心肌组织中肿瘤坏死因子-α(TNF-α)和诱导型一氧化氮合酶(iNOS) mRNA表达的影响,并阐明其作用机制。方法:取纯种新西兰幼兔36只,随机分为3组:假手术组只穿线不结扎;缺血再灌注(I/R)组,结扎30 min,再灌注2 h;ISO预处理组,缺血前吸入1.1%异氟醚30 min,洗脱15 min后处理同I/R组。缺血30 min再灌注2 h后,用放射免疫法检测心肌组织中TNF-α的含量和原位杂交法检测心肌组织中iNOS mRNA表达。结果:ISO组和I/R组幼兔心肌TNF-α含量升高,与假手术组比较差异有统计学意义(P<0.05);而ISO组与I/R组比较,心肌TNF-α含量降低,差异有统计学意义(P<0.05)。ISO组和I/R组心肌组织中iNOS mRNA的表达水平明显高于假手术组(P<0.05);ISO组与I/R组比较,iNOS mRNA表达水平明显下降(P<0.05)。结论:异氟醚通过减少TNF-α的生成和iNOS mRNA的表达以减轻I/R损伤而发挥保护作用。

Abstract:

Abstract:Objective
To investigate the effects of isoflurane(ISO) preconditioning on the expressions of TNF-α and iNOS mRNA in ischemia reperfusion myocardium of  young rabbits and clarify their mechanisms.Methods 36 young rabbits were randomly divided into three groups,the rabbits in sham operation group were not occluded coronary artery;the rabbits in ischemia-reperfusion (I/R) group were occluded for 30 min and reperfused for 2 h;the rabbits in ISO  preconditioning group were inhaled 1.1% isoflurane for 30 min before ischemia,others were the same to  I/R  group after 15 min.The content of myocardial TNF-α and the expression of iNOS mRNA in myocardioum were determined 30 min after coronary occlusion and 2 h after reperfusion. Results Compared with sham operation group,the contents of myocardial TNF-α in I/R and ISO groups were significantly increased(P<0.05);the content of myocardial TNF-α in ISO group was significantly lower than that in I/R group(P<0.05).The expressions of iNOS mRNA in I/R and ISO groups were significantly higher than that in sham operation group(P<0.05),the expression of iNOS mRNA in ISO group was significantly lower than that in I/R group(P<0.05).Conclusion ISO could reduce the TNF-α content and the expression of iNOS mRNA during I/R injury,which may be one of molecular mechanisms of  ISO  preconditioning in myocardial protection.
