摘要:

目的:观察1-甲基-4-苯基吡啶离子(MPP+)诱导的SH-SY5Y细胞帕金森病(PD)模型中可溶性抗药性相关钙结合蛋白(sorcin)表达的变化,寻求参与PD多巴胺能神经元细胞神经变性的蛋白质改变,为阐明PD的发病机制提供理论依据。方法:应用1.0 mmol·L-1 MPP+处理未分化的SH-SY5Y细胞24 h,在体外建立PD细胞模型,应用荧光染色双向差异凝胶电泳(2D-DIGE)、图像分析、基质-辅助激光解析/电离-飞行时间质谱(MALDI -TOF MS)和生物信息学
的方法,对该模型的差异表达蛋白质进行研究。结果:蛋白质组学分析在对照组和MPP+处理组之间发现22个差异表达蛋白质,其中1个蛋白质点被确切的鉴定为sorcin。sorcin的表达明显上调,荧光丰度比值为1.97倍。该蛋白的最小肽片段覆盖率、最大概率期望值、相对分子质量和等电点分别为25.7、0.006、21 940和5.3。结论:sorcin在功能上与钙离子稳态和凋亡有关,可能参与了MPP+在SH-SY5Y细胞诱导的毒性,并可能与PD的发病机制有关。

Abstract:

Abstract:ObjectiveTo observe the expression of  soluble resistance-related calcium binding protein(sorcin) in the model of Parkinson’s disease (PD) induced by 1-methyl-4-phenyl-pyridinium ion (MPP+) in SH-SY5Y cells,in order to identify proteins related to degeneration of dopaminergic neuron for revealing the potential pathogenesis of PD.Methods The undifferentiated SH-SY5Y cells were treated with 1.0 mmol·L-1 MPP+ for 24 h to set up in vitro PD cell models.Then,quantitative two-dimensional difference in-gel electrophoresis (2D-DIGE) coupled with matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) was performed to identify the differentially expressed proteins in PD models.Results Proteomics analysis revealed that 22 proteins were differentially altered in MPP+-treated SH-SY5Y cells,of which one protein spot was nambiguously identified as sorcin.Sorcin showed a significant increase in level in this PD model,and the fold changes in protein expression between MPP+-treated SH-SY5Y cells and controls was 1.97 (P<0.01).Its coverage,expectation and theoretical Mr and pI was 25.7,0.006,21 940 and 5.3,respectively.Conclusion
The function role of sorcin is correlated with calcium homeostasis and apoptosis,which may be pertinent in the toxicity induced by MPP+ in SH-SY5Y cells and  underlying pathogenesis of PD.
