多种细胞因子在CD34+细胞增殖分化中的作用

摘要: 目的:探讨血小板生成素(TPO)等多种细胞因子在分离后的脐血造血干/祖细胞体外扩增中的作用。方法:在集落培养体系及体外液体扩增体系中,分别用含有和不含有TPO细胞因子组合实验,比较其结果。结果:含有TPO组其诱导红系爆式集落单位(BFU-E )和粒巨细胞集落形成单位(GFU-GM)集落形成倍数、细胞总数和CD34+细胞增殖倍数明显高于不含有TPO组。 结论:TPO与干细胞因子(SCF)、白细胞介素3(IL-3)等其他细胞因子协同可明显提高早期造血干/祖细胞扩增效率。

Abstract: Objective To study thrombopoietin (TPO) in the expansion of cord blood hematopoietic stem/projenitor cells in vitro. Methods In the colony culture system and the liquid expansion in vitro, different cytokines combination groups with or without TPO were used to observe the result. Results The proliferation folds of colony BFU-E and colony CFU-GM, the total cells and CD34+ cells in the groups with TPO were in obviously higher than those in the groups without TPO (P<0.05). Conclusion TPO can improve the expansion effect of the early hematopoietic stem/projenitor cells obviously with SCF,IL-3 and other traditional cytokines.
