HIF-2α在非小细胞肺癌组织中的表达及其与微血管密度、Ki67和GST-π的关系

Expression of HIF-2αin non-small cell lung cancer tissue and its relationships with MVD,Ki67,and GST-π

摘要:

目的:探讨缺氧诱导因子2α(HIF-2α)在非小细胞肺癌(NSCLC)组织中的表达,分析其与血管生成、细胞增殖和化疗耐药的关系。方法:选取112例NSCLC患者癌组织和20名正常人肺组织,采用免疫组织化学EnVision法检测癌组织和正常肺组织中HIF-2α、CD31、Ki67和GST-π的表达,分析HIF-2α表达与微血管密度(MVD)、Ki67和GST-π表达的关系。结果:NSCLC组织中HIF-2α阳性表达率显著高于正常肺组织(P<0.05),在112例NSCLC标本中HIF-2α表达率为47.3%(53/112)。HIF-2α蛋白高表达NSCLC组MVD为31.1±14.7,明显高于HIF-2α蛋白低表达NSCLC组(24.3±15.8),差异有统计学意义(P<0.05)。在HIF-2α高表达组,Ki67高表达者占54.7%(29/53),明显高于HIF-2α蛋白低表达组(27.1%,16/59),差异有统计学意义(r=0.281,P=0.003)。HIF-2α高表达与GST-π表达无明显相关性(r=0.122,P=0.202)。结论:HIF-2α可能通过促进NSCLC的血管生成、增强肿瘤细胞的增殖能力在NSCLC发生发展中起重要作用,而其与化疗耐药可能无关。

Abstract:

Objective: To investigate the expression of hypoxia-inducible factor-2α(HIF-2α)in non-small cell lung cancer (NSCLC) tissue, and to analyze its relationships with angiogenesis,cell proliferation and chemotherapy resistance. Methods: Total 112 cases of NSCLC and 20 cases of normal lung tissues were selected, immunohistochemical method was used to detect the expressions of HIF-2α,CD31,Ki67 and GST-π in 112 cases of cancer tissue and 20 cases of normal lung tissue,and the correlations of HIF-2α expression with microvessel density (MVD),Ki67,and GST-π were analyzed. Results: The positive expression rate of HIF-2α in NSCLC tissue was significantly higher than that in normal lung tissue(P<0.05),the expression rate of HIF-2α in 112 cases of NSCLC was 47.3%(53/112). The MVD in HIF-2α protein high expression NSCLC group (31.1±14.7) was higher than that in HIF-2α protein low expression NSCLC group(24.3±15.8)(P<0.05). The cases of high expression of Ki67 in HIF-2α high expression group occupied 54.7% (29/53), and it was higher than that in HIF-2α low expression group(16/59,27.1%);there was significant difference(r=0.281,P=0.003).The high expression of HIF-2α had no obvious correlation with the expression of GST-π(r=0.122,P=0.202). Conclusion: HIF-2α may play an important role in the carcinogenesis and development of NSCLC by promoting the angiogenesis and enhancing the cell proliferation of NSCLC, but it may have no correlation with chemotherapy resistance.
