蝎毒对B16黑色素瘤细胞生长的抑制作用

Inhibitory effects of scorpion venom and its separatedcomponents on growth of B16 melanoma cells

摘要: 目的:观察蝎毒(scorpion venom, SV)及其分离组分对小鼠B16黑色素瘤细胞生长的抑制作用。方法:采用MTT法检测肿瘤细胞存活率,集落形成法检测肿瘤细胞增殖, 采用流式细胞术检测细胞周期的变化。结果:400和800 mg•L-1的SV可明显抑制B16黑色素瘤细胞生长 (P<0.01或P<0.05)。蝎毒组分(SV-Ⅰ、SV-Ⅱ 和SV-Ⅲ)在浓度为200、400 及800 mg•L-1时,对B16细胞生长有明显的抑制作用 (P<0.05~P<0.01);在400和800 mg•L-1 时,可使G0/G1期细胞数明显减少 (P<0.05~P<0.01),G2+M期细胞明显增多(P<0.05~P<0.01),S期细胞呈降低趋势,而SV对细胞周期进程未见明显影响。当SV、SV-Ⅰ、SV-Ⅱ及SV-Ⅲ的浓度为400和800 mg•L-1时, 对B16细胞集落形成均有明显的抑制作用 (P<0.01)。结论:在一定浓度范围内蝎毒及其组分对B16黑色素瘤细胞的存活与增殖均有明显的抑制作用,纯化后各组分的抑瘤作用优于蝎毒,但其抑制作用低于阳性对照丝裂霉素 (Mitomycin-c, MMC) 组。提示蝎毒引起细胞周期进程改变可能是其抑瘤作用的机制之一。

Abstract: Objective To investigate the inhibitory effects of scorpion venom (SV) and its separated components (SV-Ⅰ、SV-Ⅱ and SV-Ⅲ) on the growth of B16 melanoma cells. Methods MTT method was used to detect the survival of tumor cells treated with SV and its components in vitro and colony forming assay was applied to measure the proliferation of tumor cells. Moreover, flow cytometry was further adopted to observe the changes of cell cycle of tumor cells. Results SV-Ⅰ, SV-Ⅱ and SV-Ⅲ could inhibit the growth of B16 melanoma cells significantly at the concentration of 200, 400 and 800 mg•L-1 (P<0.05~P<0.01) and SV could inhibit the growth of B16 melanoma cells at 400 and 800 mg•L-1 (P<0.01~P<0.05). Furthermore, SV and its components could inhibit colony forming of B16 melanoma cells (P<0.01) at 400 and 800 mg•L-1. Cells in G0/G1 stage were decreased and cells in G2+M stage were increased obviously (P<0.05~P<0.01) after SV-Ⅰ, SV-Ⅱ and SV-Ⅲ were given at 400 and 800 mg•L-1, respectively. Conclusion SV and its components can inhibit the proliferation and survival of B16 melanoma cells. Moreover, the inhibitory effects of components are better than those of SV on B16 melanoma cells. Changes of cell cycle will be an important mechanism of inhibitory effects of SV.
