Effects of autoimmune regulator on CD4+CD25+Treg cells

Abstract:

Abstract:Objective  To observe the effects of autoimmune regulator(AIRE) on CD4+CD25+Treg cells,and explore the functions and mechanisms of   AIRE in peripheral tolerance.Methods RAW264.7 cells were transfected with pEGFPC3AIRE plasmid via lipofectin.Flow cytometry was used to determine the amount of CD4+CD25+Treg cells.  Then semi-quantitative RT-PCR was employed to detect the expressions of Foxp3 and TGF-β1  mRNA.The level of TGF-β1   was detected with ELISA kit. Results AIRE transfected RAW264.7 cells could directly or indirectly induce the generation of CD4+CD25+Treg cells and the increased expression of Foxp3 mRNA.Conclusion The AIRE expressed in the peripheral antigen presenting cells(APCs) could directly and/or indirectly induce the generation and functions of CD4+CD25+Treg cells,and probably plays a crucial role in the peripheral immune tolerance.
