Abstract:
Abstract:Objective To observe the effects of autoimmune regulator(AIRE) on CD4+CD25+Treg cells, and explore the functions and mechanisms of AIRE in peripheral tolerance. Methods RAW264.7 cells were transfected with pEGFPC3AIRE plasmid via lipofectin.Flow cytometry was used to determine the amount of CD4+CD25+Treg cells. Then semi-quantitative RT-PCR was employed to detect the expressions of Foxp3 and TGF-β1 mRNA.The level of TGF-β1 was detected with ELISA kit. Results AIRE transfected RAW264.7 cells could directly or indirectly induce the generation of CD4+CD25+Treg cells and the increased expression of Foxp3 mRNA.Conclusion The AIRE expressed in the peripheral antigen presenting cells(APCs) could directly and/or indirectly induce the generation and functions of CD4+CD25+Treg cells, and probably plays a crucial role in the peripheral immune tolerance.
Key words: autoimmune regulator, CD4+CD25+Treg cell, immune tolerance
