Influence of cyclosporine A in expression of MIP-1&alpha|mRNA in |kidney |tissue of STZ-induced diabetic rats

Abstract:

Abstract:Objective To study the influence of cyclosporin A (CsA)in the expression of macrophage inflammatory protein-1α (MIP-1α) mRNA in kidney tissue of  streptozotocin (STZ) - induced diabetic rats,and to explore the related mechanism.Methods Male SD rats were used to set up diabetes models with 50 mg/kg body weight by tailvein injection of STZ.The diabetic rats were randomly divided into 6 groups:CON (control group),DM(diabetic model),AMI(administrated with insulin), AML(administrated with CsA 1 mg/kg/d),AMM(administrated with CsA 4 mg/kg/d),AMH(administrated with CsA 8 mg/kg/d) groups(n=10).The expression levels were detected by RT-PCR.The MASSON positive staining areas and optical densities between various CsA groups were compared.Results Compared with  CON group,the expression level of  MIP-1α mRNA of rat lympthocyte in DM group was significantly increased(P<0.05 ).Compared with DM group,the MASSON positive staining areas and optical density values in CsA groups were significantly decreased(P<0.05).Conclusion The MIP-1α mRNA expression in kidney tissue of diabetic rats is increased and renal fibrosis is obvious.CsA intervention can decrease the MIP-1 mRNA expression in kidney tissue and reduce  renal fibrosis.
